Objective:To detect the expression of Peripheral blood cells metallothonein 1F (MT1F) mRNA and Vascular endothelial cell growth factor (VEGF) mRNA in Acute myeloid leukemia (AML) patients and to explore the relationship between the expression and occurrence, development and prognosis of AML. Method:The levels of MT1FmRNA and VEGFmRNA were measured with RT-real-time quantitative PCR method in Peripheral blood cells of 50 cases of adult AML patients and 20 control groups. Result:1.The MT1FmRNA and VEGFmRNA expressed in peripheral blood cells in all 50 patients and 20 normal,and the expression levels were different;2.The expression levels of MT1F mRNA (102.14±89.44) in before -chemotherapy of the AML patients group was significantly lower than the after-chemotherapy group (178.32±149.46),and lower than the control group (184.40±84.46) (P<0.05); The VEGFmRNA expression levels (102.10±100.09) in the before-chemotherapy group was significantly higher than the after-chemotherapy group (24.41±23.52) and the control group (26.00±16.71) (P< 0.05);3.The MT1 FmRNA expression levels (178.32±149.46) in the after -chemotherapy group of the AML patients were lower than the normal group (184.40±84.46),but there were no significant difference(P> 0.05); The VEGFmRNA expression levels (24.41±23.52) in the after-chemotherapy group of the AML patients were higher than the normal (26.00±16.71),two groups has no significant difference(P> 0.05); 4.The MT1 FmRNA levels in primary and relapsed states of AML group were (102.12±91.44) and (52.00±31.34),the MT1FmRNA levels in this two groups were significantly lower than the normal group (184.40±84.46) (P< 0.05),but there were no significant difference between the primary and relapsed states of AML group; The VEGFmRNA levels in primary and relapsed states of AML group were (73.71±71.34) and (199.43±127.68) (P < 0.05), the MTlFmRNA levels in this two groups were significantly higher than the control group (26.00±16.71) (P< 0.05); 5.there was no distinction between the levels of MTlFmRNA and VEGFmRNA in the invalid groups and valid groups(p>0.05); 6.The levels of MTlFmRNA had no correlation with the sex of different AML patients;7.The MTlFmRNA and VEGFmRNA had no correlation in the peripheral blood cells of AML patients(r=-0.162 P>0.05).Conclusion:l.The expression of MTlFmRNA in peripheral blood cells may be associated with the occurrence and development of AML;2.The expression levels of he MTlFmRNA and VEGFmRNA had no correlation in peripheral blood,there may be more cellular and molecular mechanisms involved in the development and regulation of the AML.
