Objective:This study was designed to establish fast and convenient mode of pregnant mice injected with amniotic fluid components,so as to observe the dynamic changes of the effect of the composition of the amniotic fluid on the coagulation of pregnant mice.And it can supply clues for the possible mechanisms of the effect of the composition of the amniotic fluid.Methods:(1)15 SD rats in late pregnancy with gestational age 21 days, were randomly divided into 3 groups:5 rats for control group (saline group),5 rats for the group after injected amniotic fluid 1 hour,5 rats for the group after injected amniotic fluid group of 2 hour. Amniotic fluid was injected into rat blood circulation respectively through tail vein. The rats were sacrificed in euthanasia after injected 1 and 2 hours respectively. (2) Indicators test:Got blood from right ventricle. Weight the liver, kidney and lung to calculate the organ coefficient. Serum was separated by centrifugal. The secretion of serum tryptase were measured by ELISA, and the international normalized ratio (PT-INR), prothrombin time (PT), activated partial thromboplastin time (APTT), thrombin time (TT), fibrinogen (FIB) were analysed by automatic biochemical analyzer.Result:(1) Compared with the control group, when the amniotic fluid was injected in pregnant mice 1,2 hours, mice lung, kidney, liver, organ coefficient increases, but the difference was not statistically significant. (2) Tryptase expression increased after the amniotic fluid composition injected.and 2 hours is higher than 1 hours.When compared with control group, difference was statistically significant (P<0.05); (3) After amniotic fluid composition injected, the pregnant rats PT, TT, APTT prolonged, while the most obvious is APTT .The difference compared with control group was statistically significant(P<0.05).Conclusion:(1).Tail vein injection is a useful way to establish a rat amniotic fluid model, and minimally invasive surgery can be taken. (2) PT, TT, APTT can be important, constant and reliable indicators for the coagulation, and the dynamic changes occurring at different times.APTT elongation is more definite in this study, suggesting intrinsic coagulation pathway plays an important role in coagulation. (3). Amount raising of tryptase in serum may be a result of intrinsic coagulation pathway activation.
