Objective:To investigate pharmacokinetics of hepatic arterial chemoembo-lization by gelatin sponge particles combined with epirubicin in a rabbit VX2 liver tumor modelMethods:Twelve successful models of VX2 transplantation liver tumor in New Zealand white rabbits was established by directly embedded VX2 cancer tissue particles into rabbits liver, which were divided into two groups randomly,HAI group (n=6):the epirubicin solution (epirubicin 10mg add 10ml of normal saline, lmg/ml)Transarterial chemotherapy alone in the hepatic artery; GSPs-TACE group(n=6):Gelatin sponge particles (20mg) and epirubicin solution (1mg/ml) full mix underwent transarterial Chemoembolization, Each rabbit with a 2mg/kg dose epirubicin。From the femoral vein blood samples at5,10,20,30,40,60,90,120 min after administration drug。killed immediately after 120min, excised tumor and peritumoral normal liver tissue, epirubicin concentrations in plasma were measured and liver specimens that were excised 120min after administration, epirubicin concentrations in tumorous andnontumorous liver tissues were measured use High performance liquid chromatography-mass spectrometry.Results:The results suggest that the epirubicin concentration of peripheral blood low GSPs-TACE groups was significantly lower than that for epirubicin-HAI groups,two groups Cmax at 5min after administration, The Cmax was 83.84±32.28μg/ml for the GSPs-TACE group,and 238.46±23.44μg/ml for the HAI group, GSPs-TACE group was significantly lower (p< 0.05).The epirubicin concentration of tumor tissue was44.49±16.80μg/g for the GSPs-TACE group, and18.32±8.30μg/g for the HAI group, epirubicin concentration for the GSPs-TACE group was significantly higher than that for the HAI group (p<0.05) epirubicin concentrations in nontumorous liver tissue for the GSPs-TACE and epirubicin-HAI groups were 10.62±8.03μg/g,5.81±1.59μg/g, respectively, and no significant intergroup differences were seen,but lower than that in the tumor tissue (p<0.05)。the AUC (μg.min/ml) at 0-120 min GSPs-TACE and epirubicin-HAI groups were 3416±799.0,11899±2717.17, respectively, GSPs-TACE group was significantly lower than that for the HAI group (p<0.05).Conclusions:1,The method of opening the abdomen and inserting tumor particle with negative pressure for establishing rabbit VX2 liver carcinoma model is simple, high success rate, little damage of animals'liver, and so on. It is possible to gain a reliable mature carcinoma animal modelfor the study of liver carcinoma therapy and imaging.2,Transcatheter arterial chemoembolization by Gelatin sponge particles combined with epirubicin more pronounced and increased local tumor concentrations in rabbit VX2 liver carcinoma,lower blood drug concentrations, and slow release of epirubicin.Therefore, it may be one of the mechanisms to obtain a good effect of the treatment of HCC...
