Objective This study was designed to investigate the influence of strontium ranelate on the bone structure and strength of biomechanics in the osteoporotic fracture of rats.Methods After ovariectomized and fixed the fractures on the proximal tibia, the 64 eligible rats were divided into treatment group (treated with strontium ranelate 625mg/kg/day) and treatment control group (lavaged with normal saline). Bone markers, RT-PCR, bone histomorphometry, X ray, micro CT and were tested to investigate the results.Results Comparing with control group, although the rate of bone healing in the sixth week showed insignificant differences, the expression of mRNA including BMP-2, VEGF, OPG and RANKL in treatment group has a high level than in controls. The RANKL/OPG ratio in the treatment group was greater than in controls. According to the bone histomorphometry and the scan of the Micro-CT, the skeletal mineralization rate, the BMD values, bone microstructure were improved obviously in the treatment group. The BMD values were increased by -3.6% vs 4.9% at 8 weeks post-treatment (P<0.05). The total bone volume values at 8 weeks were 25.6±3.1mm3 vs 39.1±4.8mm3(P<0.05).Better bone structure and bone deposition were observed in the treatment group.Conclusions In the early time after the fracture, strontium ranelate take a beneficial effects on the osteoporotic fracture healing of rats by contributing to the differentiation and proliferation of osteoblast and inhabiting the activities and numbers of osteoclast. It could accelerate the growth of fracture callus and increase callus volume, BMD without negative effects. It can be used to promote the healing of osteoporotic fractures, improve bone quality and prevent refractures.
