| Aflatoxin (AFT) is the strongest among all the toxines that are produced by molds and have carcinogenic effects. It is the product of Aspergillus fiavus and Aspergillus Parasiticus, including B1, B2, G1, G2, M1, M2, etc. It may contribute to animals' liver cancer, gastric cancer, rectal cancer, and cancers in breasts, ovary, small intestines, etc. The allowable concentration of AFT in food set by WHO is 15 ug/kg, and the allowable concentration of AFT in milk set by Chinese government is 0.5 ug/kg.In nature the main types of arsenic (As) are MMA, DMA, TMAO, Asâ…¤, Asâ…¢, AsB, AsC and AsS. Among them only MMA and DMA in organic As are relatively less toxic, while iAs like As (â…¢) and As(â…¤) are highly toxic. Therefore it is believed that the content of toxic As can be simply represented by the content of iAs. This is the hygienics basis for the testings of iAs' content.This article built the UPLC-FLR method of testing AFT M1 content in milks and milk powders by pretreatment and optimality of milks and powders; it also built the HGAFS method of testing the content of residual iAs in animal derived food. The methodology is also inspected. The methods have the advantage of rapidness, high sensitivity and selectivity.The main research work and conclusion of this article include:1. Built the UPLC-FLR method of testing AFT M1 content in milks and milk powders. The chromatography separation column is Waters Acquity UPLC BEH C18, the mobile is 18:18:64 methanol:acetonitrile: water. AFT M1 is completely separated in 5 minutes. The linear coefficient R is greater than 0.9984. The five recovery tests with the concentration of 10ppt,20ppt,50ppt, 100ppt and 200 ppt are in the range of 85.48%-94.78%. The author tested 106 samples of milk and milk powder from the whole country through this method, and the results proved that the AFT residuals in all the samples didn't exceed the standard.2. Based on GBT5009.11-2003, further refined and improved the pretreatment of the test of iAs'content in meat products, which simplified the experiment procedure and enhanced the accuracy. The author tested 40 samples of meat product from the whole country through this method, and the results proved that the As residuals in all the samples didn't exceed the standard. |
