| Objective:To investigate injury mechanism of ethylhexyl phthalate(DEHP) and its' metabolites single-ethylhexyl phthalate (MEHP) on spermatogenesis in young male Wistar rats.Methods:The 96 young male Wistar rats in 2 week after birth were randomly divided into 8 groups (12 each group). To lavage normal saline (0.9% NS 0.2ml·kg-1·d-1, feeding 3 weeks) in young male Wistar rats as a negative control group (NC) and cyclophosphamide (CTX 100mg·kg-1·d-1, feeding 1 week) as a postive control group(PC). To lavage respectively DEHP and MEHP at low dose group (100mg·kg-1·d-1, feeding 3 weeks), middle dose group (200mgkg·1·d-1, feeding 2 weeks), high dose group (300mg·kg-1·d-1, feeding 1 week)in the rest groups.To examine testicular tissue changes in different times and in different doses. Whats'more, to examine transforming growth factor-β1 (TGF-β1) in testicular tissue by RT-PCR,the telomerase activation by ELISA, and count sperm head and its'abnormity ration by light microscope.Results:(1),The atrophy,development retardation and amount decreasing of spermatogonia in medication groups and vacuoles and engorge of its'mitochondria whereas there is no significant difference between spermatogonia injury of high dose medication group in short time and low dose in long time.(2),The amount of sperm head was decreasing in medication groups whereas the abnormity ration of sperm head was increasing (P<0.05), also between high dose medication group in short time and low dose in long time(P> 0.05). (3),Transforming growth factor-β1 (TGF-β1) was increasing in medication groups (P<0.05),also between high dose medication group in short time and low dose in long time(P> 0.05).(4),To compared with the control group, the telomerase activation was decreasing in medication groups (P<0.05), also between high dose medication group in short time and low dose in long time(P> 0.05).Conclusion:The DEHP and its metabolites (MEHP) may markedly induce injury in spermatogenesis in young male rats. And the injury mechanism of spermatogenesis was concerned with expression increasing of TGF-β1 and decreasing of telomerase activation in lavageing DEHP and its metabolites (MEHP) in young male Wistar rats. |
