Objective: To establish the animal models of postoperative ileus and investigate the possible mechanism of postoperative ileus and the protective effect of traditional Chinese medicine Fuan(腑安).Methods: The animal model of postoperative ileus was established by laparotomy in mice. By measuring the inflammatory factors such as IL-6 and TNF-α,we investigated the mechanism of postoperative ileus after abdominal operation and the protective effect of traditional Chinese medicine Fuan.45 mices were randomly divided into normal group (n=15), NS group (n=15) and Fuan group (n=15). 2 hours after surgery, mices of NS group and Fuan group were perfused saline(0.3ml) and Fuan granule(0.3ml). The normal group would not be operated after anesthesia.24h after surgery, the mices were sacrificed, then gastric emptying rates, small bowel propulsion rate, serum IL-6 and TNF-αlevel were measured.Results:1,After laparotomy of mices,the gastric emptying rates and small bowel propulsion rate were reduced significantly in NS group, comparing to the normal group(P<0.05).And the gastric emptying rates and small bowel propulsion rate were improved significantly in Fuan group, comparing to the NS group(P<0.05).2,After laparotomy of mices,the serum IL-6 and TNF-αlevel increased significantly in NS group, comparing to the normal group(P<0.05).And the serum IL-6 and TNF-αlevel were improved significantly in Fuan group, comparing to the NS group(P<0.05).3,The gastric emptying rates and small bowel propulsion rate were negatively correlated with the serum IL-6 and TNF-αlevel. Conclusion: The data demonstrate that surgical manipulation can result in significantly inhibition of gastrointestinal motility and significantly increase of the serum IL-6 and TNF-αlevel. Administration of Fuan after surgery may prevent surgery-induced ileus through the effect on the inflammation caused by surgery.
