Objective: To study the impact of different concentrations of urokinase dissolve the hematoma in rabbit brain on the expression of AQP-4 around hematoma and the significance after experimental intracerebral hemorrhage model.Method: This experiment used 50 New Zealand rabbits, both male and female, weight 2.5-3.0 kg. The rabbit intracerebral hemorrhage models were made by injection of 0.5ml autologous whole blood. Rabbits were random performed 5 groups, every group included 10 rabbits: group A was control group, injected 0.1ml normal saline, group B to group E, rabbits were injected different concentration UK into the hematoma, the concentration of UK included 5000U/ml, 10000U/ml, 30000U/ml, 50000U/ml. Evaluated the water content of the brains and the expression of the AQP-4.Results: Compared with the control group, the brain water content was decrease significantly (P<0.01). With the concentration of the UK increased, the brain water content decreased firstly and then increased, and the brain water content of group 30000U/ml was the lowest. 30000U/ml group has a significantly difference compared with the other groups(P<0.01). Compared with the control group, the expression of the positive cell of the AQP-4 decreased (P<0.01). With the concentrations of UK increased, the expression of the positive cell of the AQP-4 decreased firstly and then increased, the lowest expression was the 30000U/ml group, and it has a significantly difference compared with the other groups(P<0.01). There was no significantly difference of the brain water content and the positive cell of the AQP-4 among the 5000U/ml, 10000U/ml and 50000U/ml groups.Conclusion: 1. In the experimental rabbit intracerebral hemorrhage models made by injected of 0.5ml autologous whole blood, the concentration of UK was 50000U/ml can increase the AQP-4 positive expression around hematoma, brain water content also increased. 2. In the experimental rabbit intracerebral hemorrhage models made by injected of 0.5ml autologous whole blood, when the concentration of UK was 30000U/ml, the brain water content and the AQP-4 positive expression was the lowest.
