Objective:To study the expression and correlation of DNA-PKcs and Ku70 in epithelial ovarian tumor tissues, and estimate the significance of the expression of them in development and progression of the epithelial ovarian tumor.Methods:This study included 3 groups of patients treated in the Department of Obstetrics and Gynecology, Liaocheng People's Hospital, between January 2006 and December 2010. Formalin-fixed paraffin sections of 52 cases of ovarian carcinoma,40 cases of benign tumors and14 cases of ovarian borderline tumors were studied by immunohistochemical technique for DNA-PKcs and Ku70. All of the patients of ovarian tumor were confirmed by operations and pathological examinations, there was no prophylactic chemotherapy or other therapy performed. The purpose of this study was to examine the expression of DNA-PKcs and Ku70 in epithelial ovarian tumor, and analyze the correlation between the expression of them and histological type,surgical-pathologic staging, pathologic grade, ascites and lymph node metastasis of the ovarian carcinoma. The positive control slides were obtained from the provider of the antibodies, using streptavidin-peroxidase(S-P) method. X2 test and spearman were performed using the statistical software package, SPSS 13.0. Significance was considered at the 0.05 level.Result:1. The positive staining of DNA-PKcs was localized in the nucleus, and the positive rate in benign ovarian epithelial tumor (OET), borderline OET and ovarian epithelial carcinoma(OEC) were 10%,35.7% and 61.5%, respectively. The positive rate of DNA-PKcs in the OEC group and borderline ovarian tumor group were significantly higher than that in the benign OET group (p<0.05), while there was no significant difference between the borderline OET group and the OEC group (p>0.05). In the OEC group, the positive rate of DNA-PKcs exhibited no significant difference between the ovary serous and mucous cystoadenocarcinoma (62.5% vs 60.0%), suggesting that the expression level of DNA-PKcs was not correlated with the pathological types. The positive rate of DNA-PKcs in G1-2 OEC is 51.4%, which was evidently lower than G3 stage (p<0.05), and early-stage (stageâ… andâ…¡) (42.1%) OEO expressed obviously lower level of DNA-PKcs than late-stage (stageâ…¢andâ…£) (72.7%) (p<0.05), suggesting expression of DNA-PKcs was closely related to pathological grade and surgical staging. The positive rate of DNArPKcs in the OEC absence of ascetic fluid was 54.6 %,and that in the OEC accompanied by ascetic fluid with and without metastatic tumor cells were 65.2%,62.5%, respectively. There was no significant difference of DNA-PKcs among the above three groups (p>0.05). Similarly, there was no significant difference between lymph node metastases group(64.7%) and lymph node metastases-negative group (60.0 %) (p>0.05).2. The positive staining of Ku70 also was localized in the nucleus, and the percentages of Ku70 positive cells in benign OET, borderline OET as well as OEC were 22.5%,42.7% and 78.8%, respectively. The percentage of Ku70 positive cells in OEC was obviously higher than that in benign OET or that in borderline OET (p<0.05), while there was no significant difference between the percentage of Ku70 positive cells in benign OET and that in borderline OET (p>0.05). There was no significant difference between the ovary serous and mucous cystoadenocarcinoma (81.3% vs 75.0%) (p>0.05). The percentage of Ku70 positive cells in early-stage (stageâ… andâ…¡) (63.2%) of the OEC was obviously lower than that in late-stage (stageâ…¢andâ…£) (87.9%) (p<0.05). The percentage of Ku70 positive cells in G1-2 (71.4%) was obviously lower than that in G3 (94.1%) (p <0.05). The percentage of Ku70 positive cells in the OEC absence of ascetic fluid (46.2%)was significantly lower than that in the OEC accompanied by ascetic fluid but absence of metastatic tumor cells (87.0%) (p<0.01). The expression level of Ku70 was significantly higher (93.8%) in the OEC accompanied by ascetic fluid with metastatic tumor cells (93.8%). The percentage of Ku70 positive cells in the OEC with lymph node metastases (94.1%) was significantly higher than that in the OEC without lymph node metastases (71.4%) (p<0.05). The percentage of DNA-PKcs positive cells was positively correlated to the percentage of Ku70 positive cells in the OEC (r=0.278).Conclusions:The expression of DNA-PKcs and Ku70 may be closely related with the development and malignant transformation of epithelial ovarian tumor. The interaction between DNA-PKcs and Ku70 plays a coordination role in the invasion and metastasis in the development of ovarian carcinoma. DNA-PKcs and Ku70 are hope to be potential biomarkers for malignant transformation and prognosis, as well as new target therapies for this disease.
