| In this paper, based on the separation of high-performance high performance liquid chromatography (HPLC) and the sensitive detection of mass spectrometry (MS), new serials methods for the coumarins'determination in three Chinese medicine were estabilshed.This paper mainly optimized parameters of mass spectrometry and chromatography. Investigated the effect of molecular ion peak, capillary voltage, fragments ion peak and collision energy, moreover the effect of stationary phase, mobile phase, flow rate and column temperature in detail.Under the optimized conditions of mass spectrometry and chromatography analysis, HPLC-MS methods for the determination of Radix Angelicae dahuricae, Radix Glehniae, Fructus Psoraleae were established successfully. Linear range, precision, detection limit and accuracy were researched one by one.In the clinical, it is widely apply Radix Angelicae dahuricae to psoriasis and leukodermia. The coumarins is the active ingredients. A new analytical method of HPLC-MS for the coumarins in Radix Angelicae dahuricae was established. The imperatorin, isoimperatorin and Oxypeucedan-in calibration curves showed good linearity in 0.05-10.0 mg·L-1,0.05-13.0 mg·L-1 and 0.1-120.0 mg·L-1 levels with the correlation coefficients bigger than 0.999. The relative standard deviation were 2.2%,2.6% and 2.8%(c=0.5 mg·L-1, n=6).The limits of detection were 0.03 mg·L-1,0.03 mg·L-1 and 0.05 mg·L-1. The recoveries of samples were between 95% and 105%.Radix Glehniae exerts a definite analgesic action. The coumarins is the active ingredients. A new analytical method of HPLC-MS for the coumarins in Radix Glehniae was established. The bergapten and xanthotol calibration curves showed good linearity in 0.05-10.0 mg·L-1 and 0.10-172.0 mg·L-1 levels with the correlation coefficients bigger than 0.999. The relative standard deviation were 1.9% and 2.7%(c= 0.5 mg·L-1, n= 6).The limits of detection were 0.03 mg·L-and 0.05 mg·L-1. The recoveries of samples were between 95% and 105%.Fructus Psoraleae exerts a definite antitumous effect. The coumarins is the active ingredients. A new analytical method of HPLC-MS for the coumarins in Fruetus Psoraleae was established. The psoralen and psoralidin calibration curves showed good linearity in 0.10-20.0 mg·L-1and 0.10-10.0 mg·L-1 levels with the correlation coefficients bigger than 0.999. The relative standard deviation were 2.5% and 2.8%(c= 0.5 mg·L-1, n= 6).The limits of detection were 0.08 mg·L-and 0.06 mg·L-1. The recoveries of samples were between 95% and 105%. |
