| Objective This study aims to explore the antitumor mechanism of sodium cantharidateSCA,through the study of SCA inhibiting HepG2growth and producing the VEGF protein invitro.Provide a theoretical basis for the SCA as a antitumor drug on the hepatic carcinomapatients.Methods human hepatoma cells HepG2were cultured in vitro and tread with differentconcentration (0.5mg/ml,1.0mg/ml,2.0mg/ml)of SCA,the control group without drugs, then cellproliferation was examined by the method of MTT assay at24h,48h,72hseparately,calculate cellgrowth rate.Culture the cells with0.5mg/ml,1.0mg/ml,2.0mg/ml concentrations of drug for24h,the control group without drugs,then detect VEGF protein expression by immunohistohisto-Chemistry.Use SPSS17.0statistical analysis software for statistical analysis.Significant deviationwas confirmed if P0.05.Result The inhibition result for the SCA on human hepatocellular carcinoma cells HepG2show that HepG2cells were inhibited by different concentrations of SCA, optical density(OD)decreased as the concentrations of SCA incread,mean pairwise multiple comparison resultscompared as different concentration growps were statistically significant P0.05;Immunohistochemistry technology were used to identify the VEGF protein expression in humanhepatocellular carcinoma cell line HepG2aftercultured24h with different concentration(0.5mg/ml,1.0mg/ml,2.0mg/ml)of SCA,the control group without drugs,the VEGF protein in theHepG2cells showed a brown positive reaction.brown cytoplasmic granules in the0.5ug/mlconcentration significantly reduced compared with the control group, the2.0ug/ml concentrationchanges more obviously than the group of0.5ug/ml.The expression of VEGF in HepG2cellssignificantly reduced under the action of SCA,The higher the concentration of SCA,the lowerexpression of VEGF in HepG2cells. The VEGF protein expression difference was significantbetween the experimental and control groups (F=125.75èP0.05),expression different was alsosignificant between the different experimental groups (F=120.024èP0.05).Conclusion Sodium cantharidate can inhibit the proliferation of human hepatocellularcarcinoma cells HepG2,the inhibition became more obvious as the concentration of SCAincreases and the time of action goes,the time-dosage dependent relationship existed in theinhibition of HepG2cells.SCA inhibit the VEGF expression in HepG2cells,the inhibitionbecame more obvious as the concentration of SCA goes higher. |
