Objectives:To investigate the expression of S100A6mRNA in eutopic endometrial stromalcells of OEMs while explore this kind of cells' ability of proliferation, apoptosis andinvasion. And then analysis the correlation of S100A6with the biologicalcharacteristics of this cell. In oder to discover the relationship between S100A6andOEMs.Methods:Culture and identify eutopic endometrial stromal cells in32cases of patientswith OEMs and32cases of patients with benign ovarial teratoma. Apply methylthiazolyl tetrazolium methods, flow cytometric and transwell methods to detect thiscells' capacity of proliferation, apoptosis and invasion while apply real time RT-PCRto detect the expression of S100A6mRNA.Results:We have cultured eutopic endometrial stromal cells successfully and appraisedthe purity of this cell more than90%.The0hour's OD of OEMs group(0.127±0.013)was similar with control group(0.129±0.008)(P>0.05),while the growth of24hour'sOD of OEMs group(0.148±0.020) was significantly more than control group(0.048±0.008)(P<0.01), and the growth of48hour's OD of OEMs group(0.397±0.029) was significantly more than control group(0.119±0.022)(P<0.01). Theapoptosis rate of eutopic endometrial stromal cells in OEMs group was26.43±3.79%and35.57±4.49%in control group, which reached statistical difference(P<0.01). Afterthe eutopic endometrial stromal cells got through the small room, the OD in OEMsgroup(0.950±0.014)was significantly more than control group(0.653±0.028)(P<0.01). The S100A6mRNA expressed both in OEMs group and control groupwhile the expression of S100A6mRNA was higher in OEMs group(2.908±1.413)than in control group(0.985±0.742)(P<0.01). The correlation coefficient ofS100A6mRNA and OD of proliferation in OEMs group and control group was0.947 (P<0.01),0.820(P<0.01). The correlation coefficient of S100A6mRNA andapoptosis rate in OEMs group and control group was-0.939(P<0.01),-0.800(P<0.01). The correlation coefficient of S100A6mRNA and OD of invasion inOEMs group and control group was0.967(P<0.01),0.904(P<0.01).Conclusions:The proliferation and invasion capacity of eutopic endometrial stromal cells inOEMs group was more powerful than in control group while the expression ofS100A6mRNA was more in OEMs group. However the apoptosis ability of eutopicendometrial stromal cells in OEMs group was weeker than control group. S100A6mRNA in OEMs group and control group had a positive correlation with eutopicendometrial stromal cells' ability of proliferation and invasion while had a negativecorrelation with this cells' ability of apoptosis. High S100A6mRNA expression wasprobably related with the biological characteristics of eutopic endometrial stromalcells of OEMs and might be involved in the occurrance and development of OEMs....
