Objective:To compare the expression of aquaporin(AQP) in lung tissue of hemorrhagic shock dogs with quantitative infusion directive therapy, central venous pressure directive therapy, goal directed fluid therapy in order to probe into the effects of goal directed fluid therapy to the expression of AQP in lung tissue of hemorrhagic shock dogs. Method:Eighteen heathy adult male beagle dogs were randomly divided into three groups:GDT group(experimental group,n=6), CVP group(control group,n=6), Q group(control group,n=6). The hemorrhagic shock model were established with the modified Wigger's method:the dogs were made to bleed from the femoral artery till the mean artery pressure (MAP) declined to one half of basic MAP within15minutes. This situation was maintained up to60minutes. Then three kind of fluid resuscitation was made to them and observation for4hours. After that, lung tissue was sampled to measure values of wet-to-dry lung weight ratio(W/D), and immunohistochemitry used for detection of expression of AQP1and AQP5, and HE stain used to observe pathological changes of lung tissue. Results (1). Compared with the Q and C groups, the G group significantly increased wet-to-dry lung weight ratio and the amount of infusion.(2). AQP1and AQP5were strongly positive expression in the Q and C groups, the G group reduced the expression of AQP1and AQP5.(3). In the light microscope, the lung tissue structure of the Q and C groups is complete. The alveolar septum widened, the structure of the alveolar wall was destructed and the inflammatory cells infiltrated in the G group. Conclusion:(1)The fluid volume of goal directed fluid therapy is more than quantitative infusion directive therapy, central venous pressure directive therapy.(2)Goal directed fluid therapy will increase the water content of lung tissue.(3)Compared with quantitative infusion directive therapy and central venous pressure directive therapy, the expression of AQP in lung tissue of hemorrhagic shock dogs is lower.(4)During fluid therapy in the dog of hemorrhagic shock, AQP may involve in the transportation of lung water.
