| Background:Leprosy is a chronic diseases which caused by the mycobacterium, skin and peripheral nerve involvement in. Human beings are the main host of the mycobacterium and the source of infection. Untreated leprosy patients(mainly the multibacillary leprosy patients) can infect through the respiratory tract and skin. But under the same conditions of equal access to leprosy patients, only a few people suffered disease, this is because the leprosy patients with different degrees of cellular immune deficiency. The precise mechanisms of cells immunodeficiency is still unknown. Recently Th17cells are gradually considered as a separate helper T cells subsets,and can inhibit the activation function of the other T cells, namely immunoregulation or immunodepression. RORyt (Orphan nuclear receptor) is the transcription factor which control the differentiation of Th17cells.Studies have reported that the pathogenesis of tuberculosis had associsted with Th17cells and RORγt.Tuberculosis and leprosy have the same characteristic, so our study will discuss the relationship between leprosy with Th17cells and RORyt.Objective:In the study,we research the expression of RORyt and Thl7in the lesions of leprosy, in order to expound the role of RORyt and Th17cells in leprosy.Methods:30leprosy patients and20health peoples as controls came into our research.1,Th17cells was detected on the T cells form the slides tissues of30cases of leprosy and20controls by immunohistochemistry method.2,RORyt was detected on the T cells form the slides tissues of30cases of leprosy and20controls by immunohistochemistry method.Results:1,Th17cells in leprosy were predominantly located in dermis, which were expressed in the cytoplasm,and it was absent or scant in normal skin as control. In TT and BT, Th17cells were observed both inside and around the granulomas. In BL and LL,Th17cells were present randomly in the diffuse macrophages infiltrate. Thl7cells expression were significantly elevated in patients (particulary TT) compared to controls (p<0.001). Th17cells expression was also significantly different between LL, BL, BT and TT.2,RORyt in leprosy were predominantly located in dermis, which were expressed in the cytoplasm,and it was absent or scant in normal skin as control. In TT and BT, RORyt were observed both inside and around the granulomas. In BL and LL, RORyt were present randomly in the diffuse macrophages infiltrate. RORyt expression were significantly elevated in patients (particulary TT) compared to controls (p<0.001). RORyt expression was also significantly different between LL, BL, BT and TT.Conclusion:1,Th17cells play an important role in the pathogenesis of leprosy.2, There was a possible functional interaction between Thl7cells and histiocytes.3, Thl7cells expression were significantly elevated in patients compared to controls (p <0.001).Thl7cells gradually increase in dermis from LL to TT.Above-mentionned consequence illuminate that Thl7cells may have protective effects in leprosy patients.4,RORyt expression were significantly elevated in patients compared to controls (p <0.001),which prompt that the pathogenesis of leprosy may be related with RORyt,and may play an important role by adjust the differentiation of Thl7cells. RORyt gradually increase in dermis from LL to TT.Above-mentionned consequence illuminate that RORyt may have protective effects in leprosy patients.5,Thl7cells have the func(?) to resist outside infection.At the same time,Thl7cells can cause inflammation in local and resist outside pathogen. |
