[Objective] To observe the effect of epidermal growth factor receptor inhibitors (Nimotuzumab) combined with radiotherapy inhibiting proliferation and induction of cell apoptosis on human cervical cancer cell Hela and to find out experimental feasibility of Nimotuzumab radiosensitivity.[Method] The inhibiting effect of different concentrations of Nimotuzumab on cervical cancer cell Hela was observed with colorimetric test(MTT) and the optiomal concentration was calculated.Time inhibition of Nimotuzumab combined with radiotherapy on cervical cancer cell Hela was detected with MTT and the effection to cell apoptosis and cell cycle detected by flow cytometry Annexin V/PI.[Results]1. After Nimotuzumab treated, degree of inverted microscope and cervical cancer cell Hela after treatment was found with the drugs and the role of time increased.lt was found that Hela cells became round and floating, refraction was low and loosely, cell proliferation slow down.2. MTT assay different concentrations of h-R3at different time points on Hela cells growth inhibition effect of different concentrations in the cervical cancer h-R3Hela cells, with the increase in inhibitor concentration.treatment time, tumor cell survival rate gradually decreased.3. h-R3concentration of50ug/ml, single radiation dose2Gy.the effect of combination therapy on cervical cancer Hela cells is the strongest than the drug alone or radiotherapy alone.4. Annexin V/PI double staining flow cytometry to detect apoptosis:h-R3concentration of50ug/ml, single dose2Gy. function72hours.the control group apoptosis rate was only0.06%, h-R3irradiated group apoptosis rate was9.07%.5. PI staining flow cytometry to detect cell cycle:the human cervical cancer cells Hela apoptosis rate and the percentage of G2/M phase was negatively correlate. h-R3increase the radiosensitivity of cells in G2/M phase.[Conclusion]1. Epidermal growth factor receptor inhibitor Nimotuzumab(h-R3) has radiosensitive effection on human cervical carcinoma cell Hela while combined with radiotherapy, the effect of combination therapy is the strongest than the drug alone or radiotherapy alone.,2. Epidermal growth factor receptor inhibitor Nimotuzumab (h-R3) has a inhibition of cell in the G2/M phase.
