Actinobacteria is a kind of important microorganism, which is the main source of many active natural products including antibiotics. All of bioactive substances from the microbial sources, which produced by actinomycetes have been accounted for2/3.The Amycolatopsis is an important rare Actinomycete. Until now, there are total46species in this genus validly published, among of which, there are11members can produce antibiotics or bioactive substances. The most representatives of the antibiotics are vancomycin and rifamycin. However, there are no reports about the secondary metabolites from the halophilic Amycolatopsis spp.In this thesis, four halophilic Amycolalopsis stains were involved to investigate their fermentation conditions, metal ions, chemical diversity (TLC and HPLC). By these ways, the stain YIM93223cultivated by using ISP4as basal medium. Because some metal ions can play a role on the expression of microbial secondary metabolites, using ISP4as basal medium, the YIM93223as the target strain, and add8metal ions to the medium. Then its fermentation products screened the chemical diversity by TLC and HPLC. The metal ion Mg2+to the medium was selected. Through lager fermentation, YIM93223was further studied on its secondary metabolites. Total ten compounds were isolated and purified, including two dibutyl-phthalates, two pyrimidines, four nucleotides, one N-[2-(1H-indol-3-yl)ethyl] acetamide, and one methoxy-glucose.Next, toal9halophilic Amycolatopsis stains were further screened by the TLC, HPLC and LC-MS, and strain YIM93886was selected as candidate. And then the strain YIM93886were fermented. Total9compounds were isolated and purified. Based on the data of1H NMR and some of13C NMR, that the variety of compounds produced by the YIM93886which compared with the pre-experiment is consistent. The structure of five obtained compoundsneed further analysis to confirm.
