Objective: To study the influences of modified Lianmei granules on glucose and lipid metabolism, the expression of NF-κB of liver, and the ultrastructure of liver in obesity type of2diabetes rats, explore therapeutic mechanism of modified Lianmei granules on curing type2diabetes.Methods:This experiment used48wister rats as subjects, utilizing high fat diet and streptozotocin to copy type2diabetes rat model, then randomly allocated into three groups: the model group rats(n=12), modified Lianmei granules group rats(n=12), and rosiglitazone maleate group rats(n=12). Meanwhile, the control group rats(n=12)fed common diet from the start test. After continuing to treat8weeks, blood glucose, fasting serum insulin, blood fat, and C-reactive protein were measure, while calculating insulin sensitivity indexs. Using the immunohistochemical methods the expression of NF-κB of rats's liver was determined. From cellular level to observe the change of ultrastructure of type2diabetes rat's liver by transmission electron microscope.Results:1. Contrast to the model group, modified Lianmei granules has a lower level of obesity type2diabetes rats'blood glucose, fasting serum insulin, blood fat, total cholesterol, low-density lipoprotein and C-reactive protein, a significant higher of insulin sensitivity index (P<0.01).2. Expression of NF-κB protein in the liver of diabetic rats:①The immunohistochemical results showed that the expression of rat liver's NF-κB in modified Lianmei granules group was significantly decreased to compared with that in the model group.②Semi-quantitative score displayed that the local weak staining of modified Lianmei granules group and rosiglitazone maleate group was significant difference in the model group with diffuse strong staining.3. Ultrastructure of the liver of diabetic rats shows:In hepatic sinusoids intervals of model group, the nucleus is tight, and there were rich lipid droplets within cytoplasm. Hepatocyte was observed in rosiglitazone maleate group, with a few lipid droplets, mild swelling mitochondria, small amounts of fractured rough and damaged microvilli of cell surface. In modified Lianmei granules group, injured hepatocyte was considerably reduced, approaching to normal's. The latter two groups of hepatic fatty degeneration were significantly weaker than the model group. Conclusion:1.The modified Lianmei granules has blood sugar, lipid-lowering, reducing weight and improving insulin sensitivity, reduced levels of inflammatory factors, improved the IR role.2. The expression of NF-κB protein of the rats's liver significantly increased in the model group, proved that NF-κB may be one of the important factors in liver inflammation of obesity T2DM; Through modified Lianmei granules reduced the expression of NF-κB protein in rats's liver, suggested that NF-κB be one of the target of this Chinese herbal compound.3. The ultrastructure of liver was injured seriously in model group, caused obesity T2DM may accompany with a certain of fatty degeneration of the liver, further more, correlated with T2DM and NAFLD; The results suggests that modified Lianmei granules can protect diabetic rats's liver cells, the mechanism might be related to regulate the expression of NF-κB and improved the IR role.
