| Objective: Diabetic cardiomyopathy is one of the chronic complications of diabetes.This subject of observation of Jia Wei Lian Mei Granules soup to STZ-induced T2DM rats,observe the role to reduce the levels of glucose and lipid metabolism, inflammatory factor ofplasminogen activator inhibitor-1(PAI-1) in the myocardial tissue and the influence onmyocardial tissue ultra-micro-structure change. Discussion on the relationship oninflammation of myocardial tissue with T2DM, insulin resistance deeply investigate thepathogenesis of micro-inflammation of the obese T2DM theoretical and experimental basis,and provide experimental basis for further theory of "loss drug Xinluo".Methods: The animal experiments chosed Male Wister rats a high fat diet for4weeks,and have intraperitoneal injection of STZ. Successful modeled rats were randomly dividedinto model group and Jia Wei Lian Mei Granules, maleate rosiglitazone group, each groupincluded12rats. selection the12normal rats was the blank control group. Observation theindicators of glucose, lipid metabolism and C-reactive protein; using immunohistochemicaltechniques, the expression of PAI-1in myocardial tissue in protein levels; taking theventricular tissue sections, observe the morphological changes of myocardial tissue at theelectron microscope.Results:1. In the experimental period, the model group, Jia Wei Lian Mei Granulesgroup, maleate rsiglitazone group showed no significant difference in food intake, threegroups of feeding were higher than the normal group (P<0.01); Jia Wei Lian Mei Granules13w intake lower than model group (P<0.01).2. In the experimental period, the model group, Jia Wei Lian Mei Granules group,maleate rsiglitazone group were higher in body weight comparison than the normal group(P<0.01); Jia Wei Lian Mei Granules13w in body weight comparison lower than modelgroup (P<0.01), and was significantly lower than normal rats (P<0.05).3. In the experimental period, the model group, Jia Wei Lian Mei Granules group,maleate rsiglitazone group were higher in Glucose, FIns comparison than the normal group(P<0.01), ISI was lower than the model group(P<0.01). But Jia Wei Lian Mei Granules group,maleate rsiglitazone group were significantly lower in Glucose, FIns than the model group (P<0.01), ISI was higher than the model group(P<0.01). Comparison two groups weresignificantly higher than the normal rat group, treatment group was significantly lower thanmodel group (P<0.01), but higher than the normal group (P<0.05).4. Blood lipids comparison: CHO, TG, HDL, LDL levels of model group weresignificantly increased compared with normal group (P<0.01) and were similar with theliterature. CHO, TG, HDL, LDL levels of Jia Wei Lian Mei Granules group was significantlylower than the model group (P<0.01or P<0.05), and CHO levels was significant lower thanmaleate rsiglitazone group (P<0.01). LDL levels of maleate rsiglitazone group wassignificantly decreased than the model group (P<0.01). Compared with model group, therewere not significant differences with maleate rosiglitazone in the level of CHO, TG (P>0.05).5. C-reactive protein comparison: The model group were significantly higher than othergroups (P<0.01). CRP levels of Jia Wei Lian Mei Granules group, maleate Rosiglitazonegroup were not significantly difference from the normal group.6. In diabetic model group, PAI-1expression in myocardial tissue was significantlyincreased; Jia Wei Lian Mei Granules group and maleate rosiglitazone group, PAI-1expression were significantly decreased compared with the model group.7. TEM observation: In Jia Wei Lian Mei Granules group, the chang of myocardial tissuewas close to the normal group, and was significant improvement than the model group.Conclusion: In recent year, to be widely recognized, T2DM is a kind of a chroniclow-grade inflammatory disease. IR are closely related to T2DM and its complications.Combined with traditional Chinese medicine and based on modern medical research, it usemodern research methods to discuss the therapeutic effect of traditional Chinese in T2DM andits complications. Jia Wei Lian Mei Granules have effectively theraputic in lower glucose andlipid, weight loss and improve the IR. Jia Wei Lian Mei Granules group was significantlylower PAI-1expression in myocardial tissue, be suggested that PAI-1may be a new target fortreatment of the myocardial tissue inflammatory of T2DM. |
