| Objective:To observe the effects of medium-intensity aerobic exercise and fetal liver cell transplantation on repair and regeneration cytokines of liver in rats with obstructive jaundice.Methods:Forty healthy male SD rats, weighing180-250g, were randomly divided into four groups:normal group (N group), obstructive jaundice group (OJ group), fetal liver cell transplantation group (FL group), and fetal liver cell transplantation combined with exercise group (FL exercise group). All the rats were sampled after12weeks and various target markers were determined as follows:pathological changes were observed by microscope; expression of EGF, HGE and MRP2proteins were detected with SABC; EGF, FXR and MRP2mRNA expression were determined with RT-PCR; ALT, AST and TBIL were also measured.Results:â‘ Observing the pathological changes by HE-staining, massive hepatocyte swelling, necrosis, bile duct proliferation, microvascular congestion, and fibrosis occurred in the OJ group. Tissue injury and bile duct proliferation was significantly decreased in the FL group, but not to the extent of that in the FL exercise group. Liver cable arranged normally in N group.â‘¡Serum ALT, ALT, and TBIL levels in both FL and FL exercise groups were significantly lower than in the OJ group (P<0.05), with the FL exercise group with the lowest observable levels.â‘¢HGF and EGF expression were significantly higher in both FL and FL exercise groups compared with OJ group (P<0.01), and MRP2level was significantly higher than that of OJ group (P<0.05).â‘£Compared with the OJ group, there was an increase in EGF, FXR and MRP2mRNA expression levels demonstrated in both FL and FL exercise groups (P<0.05), the EGF,FXR and MRP2mRNA expression levels was the highest in FL exercise group.Conclusion:Aerobic exercise and fetal liver cell transplantation combined with exercise play an important role in rat liver regeneration in the setting of OJ by decreasing HGF and EGF expression and elevating MRP2, the effect of fetal liver cell transplantation combined with exercise is more apparent. |
