There are many kinds of covalent modification of histone like methylation, acetylation and ubiquitinoylation, which influence the structure of chromosome, therefore have an effect on gene expression. Research showed that Arabidopsis Thaliana mutant with loss-of-function SDG8which encodes methyltransferase, resulted in changes in specific phenotype, including early flowering, lessening in plant organs, increase of stem branches and so on. This research studies the SDG8promoter dissection and hormone-induced response by cell biology, bioinformatics and molecular biology methods. The results are as follows:1. Studied on the SDG8promoter through bioinformatics method and found: trauma-related motif WRKY710S (TGAC), light regulation-related element, tissue specific expression motif GATABOX (GATA) and basic function element:TATABOX (TATA). CAATBOX(CAAT) exists in-873bp and-2706bp region, and some motifs only in-873bp to-2706bp fragment, such as AACACOREOSGLUB1、ABRELATERD1、 MYB1AT.2. Constructed a long (2706bp) promoter vector pBI-SLP::GUS and a short(873bp) promoter vector pBI-SSP::GUS of SDG8.3. Transfered the pBI-SSP::GUS and pBI-SSP::GUS successfully into arabidopsis thaliana and obtained19and18transgenetic plant containing long and short promoter fragments, respectively.4. After GUS staining and GUS activity testing with the treatment of KT、ABA、 GA3、IAA、MeJA and MeSA, the results revealed that GUS expression increases after treated with ABA, GA3, while GUS expression decreases after treated with KT, MeJA, MeSA and IAA.5. Detection of SDG8and FLC expression by quantity PCR proved that:SDG8expression increased after treated with ABA and GA3, while decreased after treated with KT、MeJA、MeSA and IAA. FLC expression increased to different degree after treated ABA、GA3、IAA、KT and MeJA, while decreased after treated with MeSA.
