Cellular Fatty Acids (CFA) is indispensable energy and nutrients of the organisms, it is one of the basic structural components of the organism. We can base on the difference of CFA kinds and contents of Bacteria to identify the strains, and analysis the phylogenetic of isolates.Bartonella is one pathogens of Zoonosis, harsh nutritional conditions, it have not been solved that the cultivation of some Bartonella. In this thesis, it through the liquid method that optimized the liquid medium composition of Bartonella, and mapped out the growth curve of11Bartonella standard strains.The experiment provided a basis for determining the extraction time and it solved the problem about culture of B.koehlerae.We select and optimize the conditions (Temperature, medium composition, passages, CO2concentration, blood kind and so on) that can affect the CFA compositions and contents of Bartonella. We constructed a standard system of Bartonella by gas chromatograph Analysis of Cellular Fatty Acids (CFA)(Culture condition:35℃,5%CO2; Medium:Trypticase Soy Agar with5%defibrinated sheep blood; Passages:3-6generations; Incubation time:4-6days; Bacteria quality:about40mg)Analysis the Bartonella standard strains and three control strains (Escherichia coli, Staphylococcus aureus, Agrobacteriumtumefaciens) by Cluster analysis and Principal Component analysis, there are very clear distinctions between Bartonella and control strains by two methods.In these three control strains, Agrobacteriumtumefaciens have very close affinity to Bartonella. Analysis the different kinds of Bartonella standard strains by Cluster analysis, that there also are clear distinctions among them.We extract the fatty acid from71wild strains by the standard system of Bartonella fatty acid that we established, then we analysis it by gas chromatography(GC) and cluster the result of GC. The results show that the CFA can be used to classify and identify the wild strains of Bartonella.
