If dry wood were amenable to molecular genetic investigations, this could lead to major applications in wood forensics, certification, archaeology and palaeobotany. The key to solving this problem lies in the extraction of the wood’s DNA and effective identification method, but the research on this has little reports in domestic and foreign countries. Therefore, to explore the wood’s DNA extraction method, and its identification methods which base on molecular level, has a certain significance.In this research, using different DNA extraction method is carried out on the DNA extraction of raw wood, and the extraction process is optimized.On this basis, different parts of the timber and wood derived from the different years are carrying out the DNA extraction, and the DNA amplification. Finally, by using DNA barcodes research standard, we had the fragments of amplification screening.The results showed that the existing DNA extraction methods can be improved for the extraction of timber’s DNA, During the process of timber’s DNA extraction, grinding, incubation time and the settling time is particularly important. Extraction of timber’s DNA in different parts shows that DNA contents in cambium, sapwood and heartwood reduce in turn, but the impurities’contents are increasing in turn. For the samples after the preservation of wood in terms of different ages, time extension with the materials out of the vitro, DNA degrades continuously. For the timber which saved for one or two years the complete genome DNA can be basically extracted. But for those woods saved more than5years the complete genome DNA were unable to extract and PCR amplification just got500bp fragment or below. For the gloomy woods which were buried in the ground for many years, PCR amplification cannot be carried out. In the screening method by using DNA barcodes, namely the genetic distance rank sum test, barcoding gap test, system tree test, indicating that the sequence of ITS2, trnL-F are more in line with the requirements of DNA barcodes and can be identified as sequence of Taxodiaceae wood.
