Among all the products of Bright bacteria, Armillariella Oral Solution, was a popular product which contained many kinds of nutrition. However, many enterprises, such as HeFei Cheng Zhi Biological Pharmaceutical Co Ltd, producted Armillariella Oral Solution through solid fermentation at present, and this fermentation system had a long production period, and easily infected by some other bacteria. So these characteristics restricted the industrialization production of Armillariella Oral Solution producted by this company. Liquid fermentation of light bacteria was still researched in the laboratory. So, the process optimization of Armillariella Oral Solution producted by this company would be helpful in the future.Based on the light bacteria as experiment strains and Medium formula which were supplied by Cheng Zhi Biological Pharmaceutical Co Ltd, the subject studied and did many researches about the production process of Armillariella Oral Solution, so as to achieve the purpose of improvement and optimization. The research content included,(1)The conditions optimization of liquid seed culture of Armillariella and the study for application of liquid strain;(2)The improvement of technology of solid fermentation condition of Armillariella;(3)The conditions optimization of extraction of Armillariella polysaccharides;(4)The research of liquid fermentation of Armillariella.Main results are as follows:(1) The research result of liquid seed:Firstly, the results of single factor test were as follows:the suitable carbon source, nitrogen source, inorganic source and dispersant were corn starch meal, yeast extract, KH2PO4and2ml/L tween80respectively according to the mycelial biomass of light bacteria; Secondly, according to the single factor experiment results, it can be found that the medium formula which was suitable for growth of mycelia was corn starch30g/L, yeast leaching powder3g/L and KH2PO41.5g/L through orthogonal test; Finally, we can learn from the test that it could enhance the output of solid fermentation metabolite obviously when vaccinated liquid seed. (2) The improvement result of technology about solid fermentation:Through adding some agricultural products, organic nitrogen source and inorganic source to the current medium and detecting the metabolites content of medium after fermentation, it can be shown that mycelia in the mediums with additives growed faster about10-15days than ordinary mediums, and the contents of metabolic product in these mediums were higher; At the same time, this subject still studied the inoculation quantity and inoculation way of liquid seed, the results showed that mycelia in the mediums with2%of the quantity, Multi-point vaccination of the liquid seed growed faster,and the contents of metabolic product in these mediums were higher.(3) The results of single factor test and orthogonal test showed that the optimum extracting conditions were as follows:the extraction time,the extraction temperature and the ratio of sample to water were90℃,1:3and2.5h. In this case, the yield of polysaccharide extraction could be6.631%.(4) The otimizing result of system factors about liquid fermentation conditions were as follows:Through the selecting tests, it could be known that sugar and com starch were suitable for liquid fermentation, the suitable nitrogen source was the mixture of typtone and yeast extract.
