It has extensive and far-reaching prospects. However, the production of Chaenomeles speciosa (Sweet) Nakai is facing many problems which greatly limited its development. Therefore, the study on biological activity and comprehensive use of Chaenomeles speciosa (Sweet) Nakai are inevitable. The dissertation studied the extraction technology and content of the effective components, purification and the analgesic and anti-inflammatory effects of the total organic acids from Chaenomeles speciosa (Sweet) Nakai. Then, the inhibition effects of Chaenomeles speciosa (Sweet) Nakai on activities of tyrosinase were studied. The main experiment contents as follows:(1) Samples were picked regularly in the same base, and the contents of total flavonoids (TF), saponins (TS), organic acids (TOA) of samples were tested. According to different process direction, the study provided a reference for choosing suitable varieties and harvesting time. The results showed that Lohan harvested in late June and Ping-guohong harvested in early August have be used in food processing. The Grafted variety harvested in July has be used in the medical and healthcare field.(2) The extraction technology of TOA, TF and TS from Chaenomeles speciosa (Sweet) Nakai was optimized by orthogonal analysis. The comprehensive extraction technology was confirmed as follows:first,75%ethanol extraction solvent,65℃extraction temperature,1:25of ratio of material to solvent,2.5h extraction time. Second,80%ethanol extraction solvent,90℃extraction temperature,1:25of ratio of material to solvent,3h extraction time. The yield of TF, TS, TOA was2.51%,1.95%,11.95%, which were higher than that in orthogonal tests. (3) The best purification of TOA was determined by comparing the purity of TOA by acid and alkali, macroporous and ion exchange resin. The results showed that the purity of TOA which purified by717GF was the highest(65.15%); the best effect of separation with the sampling volume and concentration were15.1BV and1.676mg/mL, respectively, and the elution was3.4BV0.3mol/L HC1.(4) The analgesic effect of the total organic acids and alcohol extract of Chaenomeles speciosa (Sweet) Nakai was observed by the hot-plate test and writhing analgesia test, the anti-inflammatory was observed by the method of xylene-induced mouse ear edema. The results showed that total organic acids had the analgesic effect on the pian caused by hot and acetic acid, it inhibited the inflammatory response induced by xylene and significantly reduced the ear swelling of mice. Furthermore, there was no significant difference between the total organic acids and alcohol extract of Chaenomeles speciosa (Sweet) Nakai.(5) The effects of different extract and different polarity part of Chaenomeles speciosa (Sweet) Nakai on tyrosinase activity were studied by using the Spectrophotometry. The results showed that ethanol and water extract could inhibit the activity of tyrosinase. the inhibition gradually strengthened by the concentration of sample. Furthermore, the inhibition fuction of ethanol extract was higher than water with the same concentration. The ethylacetate extract had been defined the effective part (IC50was0.148mg/mL). | | Keywords/Search Tags: | flavonoids, saponins, organic acids, extraction, purification, activities | PDF Full Text Request | Related items |
