The molecular recognition and assembly of chemical modified cyclodextrins with drug molecules have become one of the significant topic of supramolecular chemistry. To further reveal the molecular recognition mechanism and their potential application of cyclodextrins in drug delivery system, two lipophilic drug molecules, scutellarin and scutellarein were chosen for the investigation of their binding behaviors and inclusion mode with cyclodextrin by means of UV-Vis spectroscopy,1D and2D NMR, powder X-ray diffraction patterns and TG-DSC. On the other hand, some investigations on conjugation of β-cyclodextrin and scutellarin are performed. The major contents of this thesis are as follows:1. We have modified the cyclodextrin and synthesized a number of cyclodextrin derivatives. They are mono-6-O-(p-tolylsulfonyl)-β-cyclodextrin, mono(6-amino-6-deoxy)-β-cyclodextrin, mono(6-ethylene-diamino-6-deoxy)-β-cyclodextrin and mono(6-diethylene-triamino-6-deoxy)-β-cyclodextrin. Their structures are charactered by NMR spectroscopy.2. We have synthesized and chosen several cyclodextrin and cyclodextrin derivatives as host, scutellarin and scutellarein as guest. The chemistry stoichiometry and binding constant of cyclodextrins with drug molecule were investigated by Phase-solubility curves. The complex behaviors and inclusion mode are studied by UV-Vis Spetrophotometric,1HNMR spectroscopy and2D NMR spectroscopy. The characterization of solid complex was studied by means of TG-DSC, powder X-ray diffraction patterns and Scanning electron microphotographs.3. SCU-modified β-CD conjugate was synthesized from amino-modified-β-CD and Scutellarin. The structure of this conjugate was studied by NMR and MS. The characterization of solid conjugate was studied by means of TG-DSC, powder X-ray diffraction patterns and Scanning electron microphotographs.
