Based on microwave chemistry and enzymology theory, the lipase-catalyzedsynthesis of maltose fatty acid ester by eaterification had been studied,the catalyticreaction use microwave irradiation as heating method instead of conventional heatingmethod. The effects of ultrasound radiation on coconut fatty acid methyl esterspreparation by enzyme had also been studied.Compared the activities of4sorts of lipases from different sourses insynthesizing the fatty acid esters and choosed lipase Novozym435as the catalyzer onthe synthesis of maltose stearic acid ester.Using lipase Novozym435,maltose stearic acid ester had been synthesized indifferent organic solvents.The best solvent was the mixed solution of tert-butylalcohol and n-hexance(V/V=7:3).Simultaneity,maltose concentration,dosage of lipase,reaction time,reaction temperature,microwave power, molar ration of acid to alcohol,molecular sieves concentration,water content had been investigated,the optimumconditions which had been obtained through the analysis of orthogonal experimentwere as follows:70℃,maltose concentration5mmol/L,dosage of lipase20g/L,microwave power200W,molar ration of acid to alcohol3.5:1,molecular sievesconcentration70g/L,water content250μL/30mL,after reaction for26minutes,rate ofconversion could reach71.9%.The configuration of reaction products had been detected by chemicalmethods,IR sepectrum and ESI-MS.From the IR comparison chart of maltose andproduct,the absorption of1737cm-1was the absorption peak of γc=o by stretchingvibration,the absorption peak was the characteristic peak,that proved the existence ofa maltose eater.From the ESI-MS spectrogram of products,maltose monoesters andmaltose diesters were synthesized from the esterification of maltose and fat acid.Determined the nature of the maltose fatty acid ester samples,the appearance ofproducts for the powder was white or fliemot,acid value was6.7,HLB was13.4.
