Polysaccharides, which is widely distributed in animals, plants and microbescell wall, a complex biological macromolecules by the condensation of more thanvariety of monosaccharides. It is an important component of all the living organisms,which has many kinds of biological functions such as anti-tumor, immunity,hypoglycemic, anti-virus, and also has no side effects on the human body by theresearch shows. Proanthocyanidins, which is widely found in nature, is one ofpolyphenols by the condensation of a different number of monomer flavan-3-olor flavan-3,4-diol polymer. It is an efficient, low toxicity, high bioavailability ofnatural substances, it also has variety pharmacological activities such as antioxidant,anti-tumor, lower cholesterol.Tea (Camellia oLeifer Abel) seed shell, is a testa of tea seed with hard texture,mainly composed of cellulose and pentosan. The utilization of the tea seed shell wasjust focused on the manufacture of the furfural, activated carbon and xylitol. The aimof this study on the extration, seperation and purification of polysaccharide andproanthocyanidins from tea seed shell is to extend the extration resource forpolysaccharide and proanthocyanidins, and it will offer theoretical foundation tomake further use of the tea seed.Ultrasonic extraction was adopted to extract polysaccharides andproanthocyanidins from tea(Camellia oleifera Abel) seed shell, and the parameterssuch as the ultrasonic power, ratio of solid to solution, ethanol concentration,extraction temperature, extraction time which effected on the integrated extractionwere studied. In order to optimum the technology of synthetic extraction ofpolysaccharides and proanthocyanidins from tea(Camellia oLeifer) teed shell byultrasonic, the effects of ultrasonic power, ratio of solid to solution, ethanolconcentration, extraction temperature, extraction time on the extraction ofpolysaccharides and proanthocyanidins were investigated by using orthogonal designin this paper, and the factors which affected the extraction rate in the extractionprocess were analyzed by range and variance analysis. The results showed that theethanol concentration was the most distinct factors affecting the synthetic extractionyield, the ultrasonic power was the in next place, and ratio of solid to solution,extraction temperature and extraction time effects were the least, the optimumextraction conditions as follows: ultrasonic Wave300w, the ratio of solid to solution 1g:70mL,60%ethanol as solvent, extraction temperature60℃, extraction time ofultrasonic wave30min. Under this condition, the yield of polysaccharides andproanthocyanidins were0.204%and2.10%, respectively. The repeatability of theexperiments were better and the results were reliable.De-coloration experiments of crude polysaccharide by absoption resins andactivated carbon were studied. The resins HPD-200A, HPD-400A and HPD-700were selected which reprented good de-colorizing abilities and higherpolysaccharides holding rate, Besides, the de-colorizing experiments were carriedout through orthogonal experiment. The optimum technological paramteres are asfollows: HPD-700resin at30℃with30min, the de-coloration rate is20.92%, thepolysaccharide holding rate is93.36%.Chemical properties of the purified polysaccharide were analyzed, It identifiedthat the compositon of polysaccharide contained erythritol and D-fructose by theGC-MS spectra.The crude procyanidins from the tea(Camellia oLeifer) Seed Shell was isolatedby the Sephadex LH-20column chromatography, it was eluted with60%ethanol,60%acetone solution as followed, and the eluted solution with60%ethanol (termedas Sample1) and the eluted solution with60%acetone (termed as Sample2), whichwere analyzed and identificated with porter reaction, UV-scanning and HPLC. Theresults showed that Sample1and Sample2were both procyanidins, and theirrecovery were19.5mg/g and12.1mg/g, purity of them were3.8%and57.3%,respectively. and Sample1contained GA, Sample2contained EGC and otherProcyanidin monomer materials.
