Benzotriazole can be used as corrosion inhibitor, stabilizer, aircraft deicing, andantifreeze fluids. As the widely use of benzotriazole, it has been detected ingroundwater in several countries. It is reported that benzotriazole has effects on plants, plankton, and acquatic animals. However, research on effects of this chemical on theenvironment is limited. Furthermore, there is no study of toxicity of benzotriazole onmarine environment. In this study, immunotoxicity of benzotriazole was evaluated using Siganuscanaliculatus which is easily available in South China Sea and south of East ChinaSea. According to the struck of benzotriazole, end points such as HIS, enzymeactivities of GST, ACP, and AKP in serum and liver, and gene expressions of CYP1A,GST, AhR2, TNF-α, TGF-β1, and IL-1β in liver spleen and head kidney were done. The exposure time were3days and21days, and concentration of exposure were0.01mg/L,0.1mg/L, and1mg/L. The results and conclutions were as followed:1. Effect of bentriazole on liver in Siganus canaliculatus was detected, after theexposure of3-day and21-day; HIS was increased as the increasing of exposureconcentration, the result indecites that benzotriazole could induce a hyperplasia ofliver.2. Effects of bentriazole on immunity enzymes in serum and liver of Siganuscanaliculatus were detected. The activity of GST in serum was increased significantlyin the sample exposure to1mg/L for21-day; the effect of benzotriazole on activity ofACP in serum was induced firstly and then inhibited, ACP activity in liver wasinhibited; AKP activity in serum and liver was inhibited. It is means bentriazole couldlead to immune organ damage and change the pH in serum and cells in liver.3. In gene expressions test, a significant inceaseing expression of CYP1A wasobserved in liver after exposure to0.1mg/L and1mg/L for21-day and in head kidneyafter exposure to0.01mg/L and0.1mg/L for21-day; benzotriazole led an increasedexpression of GST in all the samples expect liver exposure to0.01mg/L and0.1mg/L for3-day; after3-day exposure, expression of AhR2in liver and spleen was increasedas the increasing of exposure concentration, and expression in all head kidney samplewas increased; significantly high level of expression for TNF-α was observed after1mg/L exposure for21-day, and the expression of TNF-α in spleen and head kidneywas also increased after21-day; the chemical led a significantly high expression ofTGF-β1in liver after21-day exposure to1mg/L and in head kidney after21-dayexposure to0.1mg/L; the expression of IL-1β was significantly up regulated onexposure to0.1mg/L and1mg/L for21-day. The results showed an inhibition onimmunity function by bentriazole.
