Open tubular capillary column (OTCC) is regarded as a special kind of column, which a thin layer of stationary phase (about several hundred nanometers thick) is chemically bonded or physically adsorbed onto the inner wall of capillary and the rest is open. Such unique configuration rends OTCC some desirable features such as high efficiency, low backpressure and ability for fast analysis. The purpose of this thesis is to develop the preparation technology of OTCC and its application in microscale chromatographic separation system.A rapid method for preparing OTCC applied for capillary ion chromatography has been presented. The silica capillary is firstly treated to introduce sulphonic groups, followed by dynamically coated quaternary ammonium latex particles (65nm dia.). The cationic latex particles are in place via electrostatic attraction force with anionic capillary wall. Since each particle has rich functional groups (-1450per particle), thus the column capacity obtained could be greatly enhanced compared to common manner by coating polyelectrolyte layer. The introduced sulphonic group is strong acid, which helps to increase the interaction with cationic latex particles, relative to extremely weak acid-SiO-existing in the silica capillary. Several manner of treating capillary wall have been quantitively compared and optimized. Anion OTCC demonstrated good separation of five anions and running stability.The above strategy can be extended to prepare reverse phase OTC for capillary electrochromatography. The procedure is as follows. Firstly nano-scale latex particles with dual functional groups of quaternary ammonium and C10were synthesized. The OTCC can be obtained by dynamically coating the particles solution through the silica capillary with sulphonic group treatment. The cationic latex particles are in the place via electrostatic attraction force with anionic capillary wall to introduce C10reverse phase functional groups. The influence of buffer to EOF and the repeatability and stability of the columns were discussed. The OT-CEC separated three kinds of polycyclic aromatic hydrocarbon mixtures successfully.
