Camphor tree plants in southeast and southwest of China. The trunk of Camphor tree is low and broad crown, and the shape is very beautiful. It can not only absorb a variety of toxic gases such as S, F, but also think as sand prevention, sterilization, insecticide. So it can be used to green environment. The leaves of Camphor tree with dispelling cold and relieving pain activities helps rescue various diseases. The thesis is about using different ways to extract polyphenols from Camphor tree leaves, such as microwave technology, ultrasonic wave technology, subcritical technology. Then used macroporous resin and HPLC to separate and purify extractions respectively. This experiment destination is to choose a appropriate method extracting Camphor tree polyphenols.The microwave extraction was studied to extract polyphenols from dry Cinnamanum camphoral L leaf, and to determine the optimum conditions with orthogonal test. When microwave temperature was80℃, ethanol concentration was80%, ration of material to liquid was1:60, microwave time was6min, the extraction rate of polyphenols was best,54.1mg/g.The experiment studied polyphenols from Cinnamanum camphoral L leaf with ultrasonic wave extraction technology. Some factors which influenced extraction of polyphenols such as ultrasonic time, ethanol concentration,ration of material to liquid was researched with response surface methodology. Finally, the maximum was40.8mg/g as ultrasonic time was25min, ethanol concentration was60%, ration of material to liquid was1:40.The study investigated Subcritical extraction of essential oils with N-butane, and determined the optimum conditions with orthogonal test. The factors that influenced extraction results was extraction time, extraction temperature, resolution temperature. Ultimately, the maximum was0.501%when extraction time was40min, extraction temperature was65℃, resolution temperature was45℃. The chemical composition of extracted oil was analyzed by gas chromatography-mass spectrometry (GC-MS). The analysis showed that58peaks were separated and47of them were identified. There were14olefins,8alcohols,2ketones,10alkanes,3Naphthalenes,4acids and the main constitutes were eucalyptol (24.74%)6-Octadien-3-oL,3,7-dimethyl-(5.82%), caryophyllene (4.75%), n-Hexadecanoic acid (8.55%),9,12-Octadecadienoic acid (Z,Z)-(4.30%) ect.The X-5, AB-8, NKA-II, NKA-9, HP-20, AB-8macroporous resin was used to purify extracts coming from microwave extraction technology. Finally, the AB-8was the best macroporous resin to pure Camphor polyphenols. The optimum absorption conditions of macro-porous resin AB-8were concentration of temperature30℃, pH5, ethanol concentration60%, and the absorption ratio and desorption ratio were48.24%,95.67%,separately. The factor of dynamic adsorption was feeding1.7mg/L and feeding rate was3BV/h. At last, the purity of Camphor polyphenols was61.62%after static adsorption and dynamic adsorption. |
