Study On Preparation Of Dietary Fiber From Malusasiatica Pomace By Fermentation Method And Its Functional Properties

In the paper, malusasiatica pomace were used as raw materials, preparation of dietary fiber by mixed germs fermentation method was studied, meanwhile, the dietary fiber’s physical and chemical characterisitics and relaxing bowel activity were studied also.The mainly results were as follows:1. First of all, the optimal preparation conditions of dietary fiber (DF) were determined by single germs as follow: Rhizopus oryzae, the material liquid ratio was 1:20, the inoculating amount was 8%, the temperature was 30℃, the time was 3d, the productivity of SDF of SDF was 12.20%. Lactobacillus sp., the material liquid ratio was 1:20, the inoculating amount was 10%, the temperature was 37℃, the time was 20h, the productivity of SDF was 10.62%. Saccharomyces cerevisiae, the material liquid ratio was 1:20, the inoculating amount was 12%, the temperature was 32℃, the time was 24h, the productivity of SDF was 9.58%. And then, after the testing, the mixed germs of Rhizopus oryzae, Lactobacillus sp.and Saccharomyces cerevisiae inoculation proportion was 2 to 2 to 1. Final, the optimal conditions of conditions of DF were determined by mixed germs as follow: the material liquid ratio was 1:23, the inoculating amount was 12%, the temperature was 32℃, the time was 3d, the productivity of SDF was 13.28%.2. Dietary fiber’s physical and chemical characterisitics as follow: The SWC of fermentation products at 25℃and 37℃throughed 20 mesh sieve were respectively: 4.47,4.53mL/g, and at 25℃and 37℃throughed 80 mesh sieve were respectively: 4.18,4.32mL/g.The WHC of fermentation products throughed 20 and 80 mesh sieve were respectively: 6.54,7.32g/g. The adsorption capacity on cholesterol of fermentation products was depended on acidity.At pH 7.0, its adsorption capacity on cholesterol was 7.6mg/g, the value was bigger than the at pH 2.0, its adsorption capacity on cholesterol was 5.8mg/g.The adsorption capacity on NO2-of fermentation products was still depended on acidity. At pH 2.0,its adsorption capacity on NO2- was 1.17mg/g,the value was higher than the at pH 7.0,its adsorption capacity on NO2- was 0.78mg/g. The adsorption capacities on Cd2+, Pb2+, Cu2+ of fermentation products were respectively: 40.4, 39.2, 36.5μmol/g. The value of SWC, WHC, adsorption capacity on cholesterol, NO2- and heavy metals of fermentation products was higher than the chemical fiber and raw materials.3. In superroxied radical assay system, fermentation products IC50 was 8.92mg/mL, while IC50 of Vc in this system wsa 0.71mg/mL. Its in hydroxyl radical assay system was 4.53mg/mL, while IC50 of Vc in this system wsa 0.49mg/mL. In DPPH system, the value of fermentation products and Vc were lower than 50%, which means its savenging effect in this syetem was not strong, while the max savenging of fermentation products max savenging reached 16.24%, the value lower than IC50 of Vc was 28.50%. Malusasiatica pomace DF was added into the oil, it may effectively inhibition against oil oxidating, its comparison with other antioxidants in antioxidating was respectinely investigated.4. The regulative functions of DF of malusasiatica pomace on relaxing bowel activity in mice were studied.The product could increase the percentage of propellant in small intestine of constipated model mice distinctly, the time of defecation of first feces and black feces were significant shorten, and the number and weight of feces within 8h and 12h were significant increased, compared with the control group.
