Walnut,also known as pecan,or Qiangtao,is very important nutritional nut. There arevery unique natural resources in Xinjiang province, and it is suitable for the growth ofwalnut breeding. Although there has been a lot of research on Xinjiang’s walnut bypredecessors, but researchs on the genetic diversity of Xinjiang walnut had not been veryin-depth. Especially molecular markers for genetic diversity study. In recent years, thewalnut resources continue to be suffered all kinds of damage, with the development of thetourist areas, overgrazing and deforestation and other man-made destruction. There aredifferent degrees atrophy on the distribution and density of the various regions of Xinjiangwalnut. The genetic resources were not effectively protected. In order to provide sometheoretical foundation for protection of Xinjiang walnut germplasm resources and rationaldevelopment and utilization, in this study, five different populations of Xinjiang walnutwere used to analysis their genetic diversity of germplasm resources by EST-SSR molecularmarker, then construct the dendrogram, and the results as follows:(1) Cluster analysis of5walnut populations by NTSYSpc2.1software. When GS=0.616,it can be clustered into three groups, and there are obvious regional in populations ofXinjiang walnut. By software POPGENE version32for analysis, the percentage ofpolymorphic loci (PPL) ranged from69.03to83.23%in five walnut populations, and theobserved number of alleles (Na) ranged from1.6903to1.83223; the average observationalleles (Na) was1.7626±0.4214; the average effective number of alleles (Ne) was1.3576±0.3473; the average Nei ’s gene diversity (h) was0.2171±0.1812; the average Shannon’sinformation index (I) was0.3355±0.2523. The genetic diversity analysis of walnutpopulations showed that, the genetic diversity within populations (Hs) was0.2171±0.0123,gene flow (Nm) was1.2017,indicating that gene flow among populations smooth.The analysis of variance for walnut populations by AMOVA software, the results showedthat differences among populations and within populations were highly significant, and thegenetic variation is mainly distributed within populations.(2) The DNA strips were recovered from the polyacrylamide gel, and sequence contrast,the results showed that consistency of the two sequences was67.15%. That means theamplified bands were effective.(3) The EST primers’ transferability was good, and they could be applied to EST-SSRstudy on other plants of Juglans.
