Objective To investigate the polymorphism of the GSTM1、GSTM3、GSTT1、GSTP1 genes and susceptibility to lung cancer in Mongolian population of Inner Mongolia Region. Methods The techniques of Allele Specific Polymerase Chain Reaction (AS-PCR)、Polymerase Chain Reaction-Restriction fragment Length Polymorphism(PCR-RFLP)and Multiplex PCR were used to analyze the genetic polymorphisms of GSTM1、GSTM3、GSTT1、GSTP1, the study contained 214 subjects of healthy Mongolian and 128 subjects of lung cancer patients of Mongolian nationality, and discuss the relation between the polymorphism of GSTM1, GSTM3, GSTT1, GSTP1 genes、smoke and susceptibility of lung cancer in Mongolian population. Results (1)In the analysis of the relationship with susceptibility to lung cancer was found that the people who carried with GSTM1 (-) genotype had a 2.264 fold increased risk of lung cancer than those who carried with GSTM1 (+) genotype(95% CI = 1.446 ~ 3.545), the Chi-square tests showed there was significant difference between the two groups(P<0.05). The Chi-square tests showed the genotypes of the GSTM3, GSTT1, GSTP1 polymorphisms had no significant difference among the lung cancer group and control group (p>0.05).(2)By analysis of lung cancer susceptibility combined with smoking status, we found that the nonsmokers who carried with GSTM1(-) and the smokers who carried with GSTM1(+)had increased risk of lung cancer than the nonsmokers who carried with GSTM1(+),OR values were 1.546 (95% CI = 0.771 ~ 3.100) and 1.154 (95% CI = 0.572 ~ 2.327), but there was no significant difference between the group(sP>0.05). The smokers who carried with GSTM1(+) had increased risk of lung cancer, and there was significant difference between the two groups(P<0.05).The smokers who carried with GSTM3(AA)or GSTM3(AB+BB)had increased risk of lung cancer than the nonsmokers who carried with GSTM3(AA),OR values were 2.517(95% CI =1.345~4.710) and 2.031(95% CI =1.054~3.913),and the Chi-square tests showed there was significant difference(P<0.05).The smokers who carried with GSTT1(+) and GSTT1(-) had increased risk of lung cancer than the nonsmokers who carried with GSTT1(+),OR values were 2.153(95% CI =1.176~3.944)and 2.062(95% CI =1.088~3.907, and the Chi-square tests showed there was significant differenc(eP<0.05). The smokers who carried with GSTP1(AA)或GSTP1(AG+GG) had increased risk of lung cancer than the nonsmokers who carried with GSTP1(AA),OR values were 1.806(95%CI =0.981~3.324)and 1.728(95%CI =0.938~3.182), and the Chi-square tests showed there was no significant difference(P>0.05). Conclusions In the analysis of the relationship with susceptibility to lung cancer was found that GSTM3 genotypes, GSTT1 (-), GSTP1 genotypes were suspicious risk factors of lung cancer. Smoking was the susceptibility factor of lung cancer. Gene analysis showed that an association with susceptibility of lung cancer between GSTM1, GSTM3, GSTT1 and GSTP1 was not found.
