Objective:Observation of total flavonoids of Hippophae rhamnoides on rat model ofvascular dementia effect,To investigate the mechanism of.Methods:Wistar rats,general level、180±20g、male and female half and half. Using highfat feed and improved permanent bilateral common carotid artery ligation (2-VO)preparation of hyperlipidemia vascular dementia rat model. Take the molding survivingrats were randomly divided into sham operation group, model group, the control group(Ginkgo biloba),the experimental group (total flavonoids of Hippophae rhamnoides in,high, middle, low dose group), According to the prescribed dose gavage administration,sham operation group was given the same amount of physiological saline. Each groupgavage for25days, Using the Morris water maze test model of spatial learning andmemory in rats; Gastric lavage is full after30days, blood lipid and blood rheologymeasurement, to decapitate,take out brain, nd separate the right cerebral corteximmediately for measuring activity of SOD、content of MDA. ELISA method for thedetection of serum TNF-α、IL-1β. Above all compare the differences among the groups.Results:1: Morris water maze test: during place navigation test, each group and shamoperation group compared, statistical significance(P<0.05). The control group, theexperimental group compared with the model group, statistically significant (P<0.05).During space navigation test,each group and sham operation group compared,statistical significance(P<0.05). Sham operation group, the control group, theexperimental group (middle,high dose group) compared with the model group,statistically significant (P<0.05).2:Blood lipid and blood rheology: The control group, the experimental group comparedwith sham operation group,statistically significant (P<0.05).The control group, theexperimental group compared with the model group, statistically significant (P<0.05).3:Brain tissue MDA content and SOD activity:Model group and sham operation groupcompared, statistically significant (P<0.05), Tips of model group increased the contentof MDA, SOD activity decreased. The control group, the experimental group comparedwith the model group, the content of MDA,statistical significance (P<0.05). Thecontrol group,the experimental group (medium,high dose group) compared with the modelgroup, The content of SOD, statistical significance (P<0.05). Experimental highdosage group compared with the control group, statistical significance (P<0.05).4:Serum inflammatory factor TNF-α、IL-1β: Model group and sham operation group compared, statistical significance (P<0.05), model rats serum inflammatory factorTNF-α、IL-1βcontent increased significantly. Compared with the model group, statisticalsignificance (P<0.05).Conclusion:TFH by improving the experimental rats behavior, hemodynamics, increaseantioxidant capacity, reduce brain free radical damage, effectively inhibit theinflammatory factors and mechanism, on rat model of vascular dementia and has bettertherapeutic effect.
