Objective:To observe the natural developing process of the cancer cachexia-inducingC26colon carcinoma in mouse.Methods:In B, D, F, H and J groups, a suspension of106murine colon26adenocarcinoma cells was inoculated s.c. into each mouse. Body weight,gastrocnemius muscle and epididymal adipose were documented. Biochemicalparameters and serum tumor necrosis factor-a (TNF-a) were evaluated. Theexpression of nuclear factor-kB (NF-kB) in tumor was detected by usingimmunohistochemistry. The expression of ATP-dependentubiquitin-proteasome proteolysis pathway genes Atrogin-1and MuRF-1ingastrocnemius muscle were detected by using real-time PCR.Results:Remarkable malnutrition and metabolic disorder occur about15days afterthe tumor inoculating in this model. Tumor-bearing mice in F groupcompared to healthy control mice in G group in15day: the final non-tumorbody weights in F group were lost significantly as compared with controlG group [(17.50±1.30)g VS (26.03±0.91)g, P<0.01)], Also, theconcentrations of serum total protein, serum albumin, serum triglycerideand TNF-α level showed significant difference between the two groups[(49.76±3.23)g/L vs (67.03±7.39)g/L, P<0.05;(14.44±1.12)g/L vs(19.80±2.45)g/L, P<0.01;(3.01±0.89)mmol/L vs (1.63±0.27)mmol/L,P<0.05;(199.11±17.64)ng/L vs (93.39±11.63)ng/L, P<0.01)]. The levels of the activating NF-kB and two genes, Atrogin-1/MAFbx and MuRF1whichencode proteins that are E3ubiquitin ligases, were also increased indifferent degree between the same two group camparison (P<0.05).Conclusion:Cancer cachexia developing from the inoculated tumour cells to the obvioussymptoms is a long process, the serum albumin may serve as an indicatorto judge whether or not cancer patient develop to cachectic status.
