| The Effects Of Dexmedetomidine On Behavior InNeuropathic Pain RatsObjective To investigate the effects of dexmedetomidine on behavior in neuropathicpain rats. Methods Thirty-two adult male SD rats were randomly divided intosham-operation group, chronic constriction injury(CCI)group,25ug/kgdexmedetomidine group,50ug/kg dexmedetomidine group, gave subcutaneousinjection dexmedetomidine or NS every day from postoperative to derivedpoint, determined mechanical withdrawal threshold(MWT) and thermal withdrawallatency(TWL) in preoperative1d,postoperative1dã€3dã€7dã€14d. Results Comparedwith sham-operation group, the MWT and TWL of CCI group have significantlydecrease in postoperative1dã€3dã€7dã€14d(p<0.05). Compared with CCI group, theMWT and TWL of dexmedetomidine group have significantly elevate inpostoperative1dã€3dã€7dã€14d(p<0.05),and high-dose group have significantly elevatethan low-dose group. Conclusions dexmedetomidine can inhibit neuropathic paininduced by CCI,and high-dose have more effective than low-dose.the effects of dexmedetomidine on CGRPimmunocompetenceof spinal cord in neuropathic pain ratsObjective To investigate the effects of dexmedetomidine on CGRPimmunocompetence of spinal cord in neuropathic pain rats. Methods Thirty-two adult male SD rats were randomly divided into sham-operationgroup,chronic constriction injury (CCI) group,25ug/kg dexmedetomidine group,50ug/kg dexmedetomidine group, gave subcutaneous injection dexmedetomidine orNS every day from postoperative to derived point, take lumber spinal cord to doimmunohistochemistry after determination of behavior. Results Compared with sham-operation group, the CGRP immunocompetence ofCCI group have significantly elevate(p<0.05). Compared with CCI group, the CGRPimmunocompetence of dexmedetomidine group have significantly decrease(p<0.05),and high-dose group have significantly decrease than low-dose group. Conclusions dexmedetomidine can inhibit CGRP immunocompetence of spinal cordinduced by CCI,and high-dose have more effective than low-dose.Real Time PCR detected the expression level ofCGRP-mRNA in the spinal cord in neuropathic pain ratsObjective To investigate the effects of dexmedetomidine on expression level ofCGRP-mRNA in the spinal cord in neuropathic pain rats. Methods Thirty-two adultmale SD rats were randomly divided into sham-operation group, chronic constrictioninjury (CCI) group,25ug/kg dexmedetomidine group,50ug/kg dexmedetomidinegroup, gave subcutaneous injection dexmedetomidine or NS every day frompostoperative to derived point, take lumber spinal cord to do Real Time PCR afterdetermination of behavior. Results Compared with sham-operation group, theexpression level of CGRP-mRNA of CCI group have significantly elevate(p<0.05).Compared with CCI group, the expression level of CGRP-mRNA ofdexmedetomidine group have significantly decrease(p<0.05),and high-dose grouphave significantly decrease than low-dose group. Conclusions dexmedetomidine caninhibit the expression level of CGRP-mRNA in the spinal cord induced byCCI,and high-dose have more effective than low-dose. |
