The effect of ambroxol on intratracheal instillation of amikacin rats were also investigated with microscopes.The role of drug ambroxol in the mucociliary system was reviewed.Methods30Wistar rats were randomly divided into two groups:the amikacin group(15Rats) and the ambroxol treated group(15Rats).The latter three groups were equally divided into five subgroups according to the time of2hour,4hour,8hour,24hour,48hour. Rats in amikacin group received intratracheal instillation of amikacin and the ambroxol treated group was pre-treated with ambroxol before the instillation of amikacin,and they were sacrificed at2ã€4ã€8ã€24ã€48h (3rats at each time point) after instillation,then all the trachea samples were observed by scanning electron microscope and pathologic changes of the lung tissue stained by hematoxylin-eosin(HE) were also performed with light microscope. And the impaired area of cilia were analyzed with Image Pro Plus6.0software.Results1.According to the electron microscope:In the amikacin group, the cilia of rats became lodging, sticked together, aligned abnormally, or/and abruption, mucous secretion and its stickness were increased;According to the light microscope:Pathological changes of the tracheal and bronchial epithelium cell appeared cilia disorder, mucus secretion, goblet cells increased and partially inflammatory cell infiltration.2.The area of impaired cilia of the amikacin group were more than the ambroxol treated group (P<0.01).3. The area of impaired cilia of the amikacin group was more than the ambroxol treated group at all the time spots(P<0.01).The destruction of ambroxol intervention group slightly reduced after intervention, the structure of cilia recover well.4.Within the amikacin group,the area of impaired cilia there were no difference between the2h and4h(P>0.05),was much more obviously at the2hã€4h than at the8h(P<0.01),and it was much more obviously at the24h than at the48h(P<0.01),.and within the ambroxol treated group there were the apparently reaction time was from the2h to the24h, it was more obviously at the4h than at the8h(P<0.01), it was more obviously at the24h than at the48h (P<0.01).5.Between the two groups, the area of impaired cilia of the amikacin group was more than the ambroxol treated group (P<0.01).The amikacin group at the2hã€4hã€8hã€24hã€48h were more than the ambroxol treated group.
