Kanamycin is one kind of aminoglycoside antibiotics, with characteristics ofbroad antifungal spectrum and strong potency. Because of its strong renal toxicity andototoxicity, its clinical application had been limited. Methods of semi-synthetic procedureand microbial conversion had been utilized to modify the chemical structure ofkanamycin, and a series of aminoglycoside antibiotics with low toxic and high efficacywere obtained. Microbial conversion had highly characteristic of regioselectivity andstereoselectivity, which can make reaction go smoothly while other chemical catalystcould not. In the research of microbial conversion of kanamycin B, five strains which canconvert kanamycin B were isolated from the sediment soils of Taiwan Strait. And a strain(FIM10-2207) with higher conversion was chosen for further studies. Taxonomy studiesproved that the strain is very similar to Micromonospora echinoaurantiaca. Studies on thecondition of converting Micromonspora FIM10-2207to kanamincy B was conducted andthe microbial conversion product had been separated and purified from the fermentationliquor. The physicochemical property, LC-MS and NMR of the spectrum analysis studyindicated that the microbial conversion product was1,3″-diacetylated kanamycin B. Itsvitro antibacterial activity was lower than that of kanamycin B and the experiment resultprovided a useful parent compound. In the future, it was expected to have structuretransformation and further pharmacodynamics activity research by chemicalsemi-synthetic method.
