【Objective】Toll-like receptors are the main receptors of natural immune andhave the important meaning to immune especially infection immune.As priorityreceptors of lipopolysaccharide,TLR4participate in cell activating and damage withLPS mediated in, and play an important role in the liver diseases including theoccurrence and development of chronic hepatitis B. However, few studies in theworld have reported whether the TLR4polymorphisms were associated with theoccurrence of chronic severe hepatitis B. In our study, the SNPs within the TLR4genes were analyzed in different groups of chronic hepatitis B and chronic severehepatitis B, then to evaluate the relationship of Toll-like receptor4polymorphismsand chronic severe hepatitis B.【Methods】Totally,180patients with chronic severe hepatitis B and200patientswith chronic hepatitis B were enrolled in this study. We extracted genomic DNA byDNA extraction kit from human peripheral blood mononuclear cells. The candidategenes’ SNPs were selected according to the database of Chinese Han population inNCBI, which contain two sites within the4th exon region of TLR4gene includingrs4986790(A/G)、 rs4986791(C/T). The software including vector NTI10andPrimerPremier5were applied to design the primer for PCR. The results wereanalyzed using real-time quantitative fluorescence PCR and then the differencesbetween the different groups were compared in genotype and allele frequency.【Results】1. Find rs4986790(A/G) site there exist three genotypes of AA,AG,GG, including gene type AA for wild type; rs4986791(C/T) site exist three genotypesof CC,CT, TT, including gene type CC for wild type;2.No statistical significance wasfound in genotype and allele frequency of rs4986790(A/G) site between chronicsevere hepatitis B and chronic hepatitis B patients (P>0.05), the OR value of allelefrequency was1.21, and the95%confidence interval was0.56to2.59. There was nostatistical significance in genotype and allele frequency of rs4986791(C/T) site between chronic severe hepatitis B and chronic hepatitis B patients (P>0.05), the ORvalue of allele frequency was1.08, and the95%confidence interval was0.70to1.68.【Conclusion】Two SNP sites within the exon region of TLR4gene includingrs4986790(A/G)、 rs4986791(C/T) exist in Chinese Han population in Fujianprovince,and have no correlation with chronic severe hepatitis B.
