The salivary glands were reported to be important human glands, which wereoral cavity and digestive tract essential exocrine glands as well as they hadendocrine function. They could maintain ingestion, digestion, speech and so on.If Salivary gland damaged even deficiency, there would be leaded to xerostomia(dry mouth)、dysphagia、dysgeusia, dental caries and so on. As a result, they hada harmful effect on the quality of patient‘s life. Tissue engineering of the salivaryglands give a hope for this problem,The method is that cells have grown on thescaffold material in vitro and repaired organs of damaged by transplantation invivo.In this study, the SD rat‘s bone marrow mesenchymal stem cells (BMSCs)were induced salivary gland acinar-like cells(SGALCs)which seeded onto thethree-dimensional scaffold to transplant in nude mice and reseach the effects.First, BMSCs were isolated from SD rat, cultured、 purification and identification in vitro,BMSCs were induced into SGALCs through the culturemedium. Then, cells seeded onto the three-dimensional scaffold which collagenwas combined with chitosan according to certain proportion. The experiment wasdivided into two groups:(1)experimental group: SGALCs seeded onto thethree-dimensional scaffold;(2) blank group: BMSCs seeded onto thethree-dimensional scaffold. Finally, cells were seeded onto the scaffolds weretransplanted in nude mice‘s subcutaneous,1W,2W, immunofluorescence andimmunochemical was used to check whether the two groups of cells were taggedmaintaining salivary gland cells phenotype and showing alpha-amylase (AMY)activity. The result showed experimental group of cells of1week,2weeks couldexpress AMY. The blank group was negative.In this study,we built a controllable artificial induction environment in vitro,SD rat‘s BMSCs successfully induced into SGALCs by the culture medium atfirst time. We successfully constructed a three-dimensional scaffolds for celladhesion. At the same time, SGALCs seeded onto three-dimensionalcollagen-chitosan scaffolds were successfully transplanted into nude mice andobtained a desired effect. Providing experimental evidences for establishingfurther the salivary glands of tissue engineering.
