Honeysuckle Flower as a kind of economic crops has already formed series of industry chains, like medicinal chains and food’s, which is a hot pint of Natural Medicinal research. The main chemical constituents are organic acids, flavonoids and naphtha class and other substances. Take H.F. as a drug for the significant antimicrobial activity, modern pharmacology study proves that organic acid (Chlorogenic acid accounted for most) and flavonoids have a closely related with antimicrobial activity. At present, the research of Chlorogenic acid is very common and vigorous growth of the flavonoids’At the same time, modern material science, statistical science and math develop rapidly, more and more inter discipline subject are emerging. All of this means a development opportunity for Traditional Chinese Medicine. This paper combining modern statistics and the mathematical model, as well as modern statistics and the mathematical model, Process control theory and electrochemical, Colloidal chemistry, materials science and chromatographic detection, discus the three stages, namely Experimental design optimization, The actual production technology research and the part of PQC:1. Ultrasonic extraction of chlorogenic acid of experimental parameters are optimized by central composite design-response surface(CCD-RS),and then,use the software called origin and design-expert to processes experimental data, and built the binomialequation:Y=1772.86-29.18X1-1.60X2-56.80X3+0.45X1X3+0.05X2X3+0.17X12+4.80E-0.03X22+3.58X32r=0.9644,take ethanol concentration for X1,the time for X2,Material liquid times with quantity for X3,chlorogenic acid content for According to this equation combine with RS. Get the optimal results theory, and then test it and find that comparing with the actual value deviation is3.6%, proved that the design optimization method can be used in the ultrasonic extraction of chromogenic acid in the experiment. And then, compared with orthogonal design Illustrated the design method in Chinese traditional medicine research has the superiority.2. Combine with Process control theory and electrochemical, Colloidal chemistry, to check the Traditional craft, for the extraction stage and purification stage some improvement are taken, form the new craft. Small experiment and amplification of the results of the study show that the experiment, in the extraction stage extraction ratio improve72.87%, in purification stage the loss rate of chlorogenic acid is8.66%,down more than20%.proved the feasibility of this technology. The specific process conditions is in the extraction stage take the pH to8.5,and then to meta-acid, To concentrate the content of solid solution for3%,put0.3%chitosan in,and Stirring3hours with a speed of20r/min.3. Built a detection method to Honeysuckle Flower flavonoids substances, namely rutin hyperin and quercetin with HF-LPME-UHPLC-MS, take HF-LPME as preparation technology. Chromatographic conditions:Waters AcquityTM。BEH C18(50mm×2.1mm,1.7μm), mobile phase:carbinol(A) and formic acid solution(B),flow rate0.2mL/min, gradient elution, Column temperature30℃.Mass spectrometry conditions:Ionization source:ESI, detection mode:full scan, Collision pool pressure:334Pa; Capillary voltage of:3kV;Cone hole voltage:30v.The results show that Linear range of rutin hyperin and quercetin respectively is:0.0227-1.195,0.0141~1.32,0.00832~1.649μg/mL; The average recovery respectively is:95.34%,95.17%,99.83%;Enrichment multiples(EM) respectively is:38、42、83,limit of detection(LOD) respectively is0.0290.0170.006μg/L. The result is:take HF-LPME as preparation technology for UHPLC-MS, improve the sensitivity EM and LOD. For detection of honeysuckle material flavonoids provides a reliable method.
