ObjectiveTo study the effects of hydrogen sulfide on the level of proteinuria, reactive oxygenspecies(ROS) and the expression of podocalyxin protein and mRNA in the kidney of diabeticnephropathy (DN) rats and its possible mechanism.MethodsSprague Dawley rats were injected with streptozotocin (STZ,65mg/kg) into abdominal cavityto establish DN model. DN rats were randomly divided into DN group (group D) and hydrogensulfide group (group H). Normal rats wers served as control (group N). Group H received dailyintraperitoneal injection of0.25mL NaHS (56μmmol·L-1·kg-1), group N and group D wereinjected with0.25mL sterile water. After8weeks of treatment, serum biochemical indicators andquantities of24hours urinary protein were measured by automatic biochemical analyzer. The levelof reactive oxygen species (ROS) was measured by laser scanning confocal microscopy after beenlabeled by fluorescent probes. Immunohistochemical and real-time quantitative polymerase chainreaction (RT-PCR) were applied to detect the protein and mRNA expression of podocalyxin.Results1. The impact of hydrogen sulfide on the renal fuction and24hours urinary protein of diabeticnephropathy rats: Compared with group N,24hours urinary protein, serum creatinine (Scr), bloodurea nitrogen (BUN) increased obviously in group D and group H. It increased more significantlyin group D than in group H (P <0.05).2. The impact of hydrogen sulfide on podocalyxin protein of diabetic nephropathy rats:Compared with group N, the expression of podocalyxin protein was significantly decreased ingroup D (P <0.05), while it decreased slightly in group H (p>0.05). Compared with group D,podocalyxin protein exprssion was higher (P <0.05).3. The impact of hydrogen sulfide on podocalyxin mRNA of diabetic nephropathy rats:Compared with group N, the mRNA expressions of podocalyxin were significantly reduced ingroup D and group H (P <0.05). It decreased more significantly in group D than in group H (P<0.05). 4. The impact of hydrogen sulfide on ROS of diabetic nephropathy rats: Compared withgroup N, the level of ROS was significantly increased in group D and group H. It increased moresignificantly in group D than in group H (P <0.05).5. Analysis of correlation: ROS and24hours urinary protein were positively correlated(r=0.546, P <0.01). The expression of podocalyxin mRNA and24hours urinary protein werenegatively correlated (r=-0.375, P <0.05). The expression of podocalyxin mRNA and ROS werenegatively correlated (r=-0.678, P<0.01).ConclusionsHydrogen sulfide can reduce the level of proteinuria of diabetic nephropathy rats, which maybe associated with reducing oxidative stree and up-regulating the expression of podocalyxin, thenplay renal protective effect on diabetic nephropathy.
