In the twenty-first Century, the combination of life science, information science andmanufacturing science became an important trend of the development of science and technology.3D Cell assembly technique is based on manufacturing theory and tissue engineering cellmicroarray technique is based on microelectronic mechanical system and sensor technique, thesetechniques provide new theories and techniques for tissue engineering, drug screening and otherfields. In this study, we introduced 3D Cell assembly technique into cell chip manufacturing andstudy the theory and technique for this combination.Firstly, we analyzed the cell assembly technique and cell chip technique, theories and methodsthat how to assemble cells on chip were also analyzed. Based on the mechanism of cell assemblytechnique and cell chip, we chosen MEA and ECIS biosensor for the chip, then the parameter,materials and process of the biosensors were studied. With these biosensors, we designed andfabricated an integrated cell chip which integrates several kinds of biosensor arrays. Results showedthat the chip is sensitive to detect the cell electrophysiology.Secondly, we displayed the 3D cell assembly machine and developed the nozzle system. Westudied and chose biological material for this work, and the ratio and compatibility of differentbiological materials were studied. After these work, we assembled the materials on the chip.Forming and curing characteristics, stability and the electrochemical detection effect were studied.Thirdly, according the structure of integrated cell chip, we designed the assembly path and founda optimum path. Then we studied the effects of forming parameters, nozzle size and crosslinkmethod on the path integral, continuity, cohesiveness and microfilament diameter. Results showedthat cell/materials can be assembled at designed sensor area accurately, the bonding between chipand cell is stable, and the diameter up to the requirements.Finally, we compared DMEM and PBS solution with cyanide iron potassium solution. Then, weassembled different cell/materials on chip. Results showed that the cells could been assembled intodesignated places on the chip, the cellular impedance increase curve of ADS cells calculated by chipanalysis system was consistent with the curve obtained by Alamar Blue assay, the integrated cellularphysiologic sensor on chip can detect changes of different cells. Construction and study of such cellchip systems will develop new technologies for cell chip and drug discovery.
