Effects Of Heterogeneous Umbilical Cord Mesenchymal Stem Cells Intramuscular Injection On The Myocardial VEGF And CTnI Expression And The Serum Concentration Of The VEGF,HGF,IGF-1and GM-CSF In Normal Wistar Rats

Objective To explore the effects of intramuscular injection of heterogeneous umbilical cord mesenchymal stem cells (UC-MSC) on expression of the myocardial VEGF, cTnl and the serum concentration of the VEGF,HGF,IGF-1and GM-CSF in normal Wistar rats.Methods A total of60male SPF Wistar rats were divided into six groups randomly, including the normal group(PBS as suspension liquid of human umbilical cord mesenchymal stem cells(HUMSC) was injected into muscle), the control group with culture medium (named the culture solution group: DMEM as the culture solution of HUMSC was injected into muscle),the supernatant group of hUC-MSC (named supernatant group:the supernatant of HUMSC was injected into muscle), the low concentration group of UC-MSC (named low concentration group:HUMSC with amount of0.25×105were injected into muscle),the moderate concentration group of UC-MSC (named moderate concentration group:HUMSC with amount of1.0×105were injected into muscle),the high concentration group of UC-MSC(named high cell concentration group:HUMSC with amount of4.0×105were injected into muscle). Each group included10rats and each rat was injected0.8ml liquid into muscle(0.2ml in each limb).Clinical observations were performed every day after intramuscular injection.Rats received a second intramuscular injection of HUMSC4weeks after first injection trials.4weeks after second cell injection trials all rats were executed. The serum concentration of vasuclar endothelial growth factor(VEGF), hepatocyte growth factor(HGF), insulin-like growth factor1(IGF-1) and granulocyte macrophage colony stimulating factor(GM-CSF) were measured with enzyme-linked immunosorbent assay kit(ELISA).The VEGF and cardiac troponin Ⅰ (cTnⅠ) expression of myocardium were detected by immunohistochemical technique.Result No alteration of the weight, the posture, the activity, the fur and the skin integrity of the rat were found, and no death of the rats during experiment was occurred. There was no significant difference on the serum concentration of the VEGF,HGF,IGF-1and GM-CSF among the groups before injection and8weeks after injection (F=0.676-0.870,0.278-0.406,0.194-0.741,0.936-1.042. p>0.05); In the same group, no statistical significant changes of the serum concentration of the VEGF,HGF,IGF-1and GM-CSF between before injection and8weeks after injection (t=-1.556-0.223,0.817-2.173,0.749-0.675,-1.214-1.109; p>0.05). Weak positive expression of the VEGF in the cytoplasm of cardiac myocytes in the normal group was observed., and strong positive expression of the cTnⅠ in the cytoplasm of cardiac myocytes in the normal group was observed. There was no significant difference of the VEGF and the cTnⅠ express in myocardium among the groups(F=0.651and1.019,p>0.05).Conclusion HUMSC or the supernatant of HUMSC intramuscular injection had no effect on the serum concentration of the VEGF,HGF,IGF-1,GM-CSF and the myocardial VEGF and cTnⅠ expression in normal Wistar rats.
