| ObjectiveRabbit bone marrow stromal cells (MSCs) transfected by bone morphogenetic protein-2(BMP-2) gene and transforming growth factor-β1(TGF-β1) gene mixed with calcium alginate to repair defects of articular cartilage of rabbit knee, then to observe the result of repairing and compare the repairation effect of single gene and double gene.MethodExtract bone marrow cells from a four-week-old New Zealand rabbit. Isolated and purified bone marrow stromal cells in vitro. Choose36grown-up New Zealand rabbits and divide them into6groups randomly. Full-thickness articular cartilage defects were made in the non-weight-bearing area of the bilateral femoral condyles in New Zealand rabbits. Transfer the different six groups complex component to the different defect place of the articular cartilage:In group A, MSCs+calciumalginate complex, which has been transfected by BMP-2and TGF-β1(Experimental Group); In group B, MSCs+calciumalginate complex, which has been transfected by BMP-2(Control Group I); In group C, MSCs+calciumalginate complex, which has been transfected by TGF-(31Control Group Ⅱ); In group D, MSCs+calciumalginate complex, which has been transfected by Empty plasmid (Control GroupⅢ); In group E, calciumalginate without cells (Control GroupⅣ); In group F, without any treatment (Control Group V). The femoral condyles were harvested at the end of12weeks, and then observe the condition of respairation of cartilage defects and make assessments of histology and immunohistochemistry. Evaluate the repairing effect by O’Driscoll’s and do statistics analysis.ResultsThe surface of the repaired tissue in group A is smooth and difficult to be distinguished from that of the normal structure directly to the eye, but immunohistochemistry result showed that there’s an accumulation expression of type II collagen. The repaired surface of Group B and C is also smooth, more or less, but the boundary with normal tissue is vague, the staining of type Ⅱ collagen showed expression to some extent. Obvious defects are found in Group D,E and F, with only a little type II collagen expression. The statistical analysis of histology scoring indicates that there is no significant difference between Group B and C (p>0.05), but all the others compared with each other showed statistical significance (P<0.05)ConclusionThe group which was implanted BMP-2+TGF-β1+MSCs is superior to single gene groups in the effect of repairing cartilage defects. |
