Objective To explore the Effect of Sufentanil Postconditioning on MDA and SOD ofMyocardial Ischemia Reperfusion Injury of Oxidation in Dogs and Relationship of the Effectwith JAK2-STAT3 Pathway.Methods Twenty- four dogs were randomly divided into four groups (n=6,each):①the controlgroup (Sham group):left anterior descending coronary artery were stringed without ligating;②ischemia-reperfusion group (I/R group): ligationof left anterior descending coronaryartery.After 30 min, blood flow was restored and lasted for 120 min;③sufentanil post-conditioninggroup (SPO group): intravenous injecting sufentanil (0.6μg/kg) 5 mins before reperfusion,others were treated in the same way of IR group;④sufentanil post-conditioning+ tyrosinekinase inhibitor AG490 group (SPO+AG490 group): intravenous injecting AG490 (1mg/kg) 5mins before reperfusion, others were treated in the same way of SPO group. At the end of 120min of reperfusion, the myocardial specimens were removed and were put into formaldehyde.The concentration of malondialdehyde(MDA)and activity of superoxide dismutase(SOD) inmyocardial tissue were observed, hematoxylin-eosin staining to observe the pathological changesof myocardium.Result1. Concentration of MDA Compared with Sham group, the concentration of MDA inmyocardial tissue were increased in I/R group and SPO group as well as SPO+AG490group.Compared with I/R group, the concentration of MDA in myocardial tissue becamedecreased in SPO group. Compared with SPO group, the concentration of MDA in myocardialtissue became increased in SPO+AG490 group.2. Activity of SOD Compared with Sham group, the activity of SOD in myocardial tissuewas decreased in I/R group and SPO group as well as SPO+AG490 group.Compared with I/Rgroup, the activity of SOD in myocardial tissue became increased in SPO group. Compared withSPO group, the activity of SOD in myocardial tissue became decreased in SPO+AG490 group.3.Pathological observation Compared with Sham group, myocardial cell’s degenerationand the morphological changes of cardiac muscle cell’s ultrastructure were significantlydamaged in IR group, SPO group and SPO+AG490 group, however, the changes ofSPO+AG490 group were more obviously than SPO group. Conclusion1. Sufentanil Postconditioning on Myocardial Ischemia /Reperfusion Injury of Oxidation canruduce concentration of MDA and increase activity of SOD, So the effect of SufentanilPostconditioning on Myocardial Ischemia /Reperfusion Injury of Oxida tion is protective .2.The mechanism of the protective effect of Sufentanil Postconditioning on MyocardialIschemia /Reperfusion Injury of Oxidation is relative with JAK2-STAT3 pathway.
