Objective:by means of observe different dose of GLP-1analogue Exendin-4influence to the expression of diabetes nephrosis rats kidney tissue SOD,8-OHdG, to explore the influence of the GLP-1analogue Exendin-4on kidney oxidative stress when diabetes nephrosis.Methods:DN experimental model were randomly divided into the treatment group, high dose and lower dose treatment groups. After a period of6weeks intervention with GLP-1analogues Exendin-4determination biochemical indicators such as blood sugar,24hours urinary protein quantitative, total cholesterol and triglycerides, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and HE dyeing to observe the kidney pathological changes, immunohistochemical dyeing to observe kidney tissue SOD,8-OHdG expression changes. Then statistics processing the experimental data.Results:after Exendin-46weeks treatment, compared with DN group, lower dose treatment group and high dose of the treatment group blood sugar,24h urine protein quantitative obviously ameliorated (P<0.05), high dose treatment group ameliorates more obvious (P<0.05).Lower dose treatment group and high dose treatment rats, kidney tissues of SOD express significant rise8-OHdG express declined obviously (P<0.05), high dose treatment rats kidney tissues expression of the change is more obvious (P<0.05).Conclusion:GLP-1analogue can reduce the degree of oxidative stress of kidney tissue when diabetic nephropathy, in order to delay the progress of diabetic nephropathy,and this kind of effect is dose depended. |
