| Objective:To investigate the possibility of repairing the cartilage defect with autologous bone marrow mesenchymal stem cells(BMSCs) combined with the â… /â…¢-collagen-membrane.Methods:The experiment was completed in Shanxi Medical University Second Affiliated Hospital of bone and cartilage tissue injury and repair laboratory from2010-8to2011-10.(1) In vitro isolation,culture and expansion of bone marrow mesenchymal stem cells from12New Zealand white rabbits(2month old).(2) BMSCs were cultured with the â… /â…¢-collagen-membrane to form BMSCs/collagen membrane compound in vitro.(3) Totally24New Zealand rabbits were assigned into2groups. Both knees full-thicknes s articular cartilage defects with3.8mm diameter and2mm depth were prepared.The defects were filled with autologous BMSCs/collagen membrane compound on the12New Zealand white rabbits as A group(transplantation group), and filled with only collagen membrane on the another12as control group.All the animals were sacrificed at8and12weeks after surgery.The repair tissue was investigated using gross, histological staining and with immunohistochemical staining for type-II collagen, and grading scales were evaluated according to O’Driscoll, Keeley and Salter grading method, statistical software SPSS13.0for statistical analysis.Result:(1) The third generation of bone marrow mesenchymal stem cells were observed: BMSCs growth and morphology,fusiform cell vortex formation and the histological staining: cells on a slide attached, arranged in the law,was stratified arrangement, cell morphology typical.(2) The cultured BMSCs can be well attached to the rough surface of collagen membrane.(3) In vivo:hostological stain showed that,in the group A(transplantation group)repair tissue mainly was hyaline-like cartilage at the8weeks,and it was close to normal hyaline cartilage at the12weeks.However in the group B(control group)the tissue mainly was fibrous tissue, at the weeks, and fiber-based cartilage for12weeks. (4) The histological grade of12weeks was different significantly from that of8weeks(P<0.001).There were also significant differences between experimental group and control group at each time intervals after operation(P<0.001).Conclusion:(1) Among the third generation of bone marrow mesenchymal stem cells in good condition, a single cell morphology, typical seed cells can be used as the organization.(2) Bone marrow mesenchymal stem cells can be attached growth in the â… /â…¢ collagen membrane surface, and the â… /â…¢ collagen membrane can be used as scaffolds in cartilage tissue engineering.(3) It is a good way to repair articular cartilage defects with autologous bone marrow mesenchymal stem cells,BMSCs-the â… /â…¢-collagen-membrane. |
