| Objective: To evaluate the change of serum levels ofsE-selectin and explore the association of the polymorphism of E-selectingene A561C with susceptibility to type2diabetic nephropathy patients.Methods: Serum level of sE-selectin was measured with enzyme-linkedimmunosorbent assay (ELISA). Serum levels of glucose, HbAlc, ureaand creatinine were measured by biochemical techniques. Genotype ofE-selectin was determined in125patients with type2diabeticnephropathy and106healthy controls by polymerase chainreaction-restriction fragment length polymorphism (PCR-RFLP)Results: The serum levels of sE-selectin in type2diabetic and type2diabetic nephropathy patients were much higher than these in healthycontrols respectively (P <0.05). Moreover, the serum levels of sE-selectinin type2diabetic nephropathy patients were much higher than those intype2diabetic patients without complications (P <0.05); Serum levelsof sE-selectin in type2diabetic nephropathy patients had positivecorrelation with serum levels of glucose, HbAlc, urea and creatinine. Thestudy also showed that E-selectin genotype frequencies of AA, AC and CCwere76.8%,18.4%and4.8%in type2diabetic nephropathy patients group,90.6%,8.5%and0.9%in control group and94.3%ã€5.7%ã€0%innomal control group, respectively. The allele frequencies of A, C were86%and14%in type2diabetic nephropathy patients group,94.8%and5.2%in control group and97.2%ã€2.8%in normal control group,respectively. There were significant differences in frequencies of allele andgenotype in E-selectin A561C polymorphism between type2diabeticnephropathy patients group and control group (P <0.05).Conclusion: The serum level of sE-selectin is related to type2diabetic nephropathy patients. There was an association betweenE-selectin A561C polymorphisms with type2diabetic nephropathypatients. Furthermore, C allele appears to be a risk factor. |
