Objective:Immunologic thrombocytopenic purpura (ITP),is a result ofplatelet immune destruction, hemorrhagic disease of peripheral blood plateletcount decrease,which is characterized by peripheral blood platelet count issignificantly reduced with bone marrow megakaryocyte development,maturation obstacles, clinically wide range of skin and mucous membraneinternal bleeding as the main performance. Its pathogenesis is still unknown.Killer cell immunoglobulin-like receptors (KIRs) is another immuno-modulatory gene family with a high diversity was found following the HLAcomplex is found. Previous studies have shown that the development of avariety of autoimmune diseases such as rheumatoid arthritis, type1diabetes,psoriatic arthritis, vasculitis and so on are associated with the of abnormalitiesKIR gene. This study was designed to explore whether the occurrence of ITPand the polymorphism of KIRs are relevant.Methods:The case group consists of60patients diagnosed with ITP byJilin University Norman Bethune No.1Hospital sdmitted in Uanuary2010toOctober2011and the control panel consists of60blood donors. The DNAgenome samples were extracted from these people, then the gene loci of KIRswere genotyped using low resolution polymerase chain reaction withsequence-specific primers (PCR-SSP) typing method. The gene loci ofKIR2DL1~KIR2DL5, KIR2DS1~KIR2DS5, KIR3DL1~KIR3DL3, KIR3DS1,KIR2DP1, KIR3DP1were examined.Results: In this study, we typed16KIR genes in60ITP cases and60healthy controls. All of the KIR genes tested were present in both patients andcontrols. Three framework genes (3DL2,3DL3,2DL4) and3DP1were detected in almost all the ITP cases and controls. In the respective comparisonof phenotypic frequencies of KIR genes between ITP patients and healthycontrol subjects revealed significantly different frequencies of KIR2DL3,KIR2DP1.Conclusions:1.The genetic variation at the KIR is related with susceptibility to ITP.2.The abnormal expression of KIR2DL3gene may be related to theoccurrence of ITP.3.KIR2DP1may play a role in the pathogenesis of ITP.
